精制冠心片中丹参脂溶性成分的HPLC含量测定

摘　　要：目的建立精制冠心片中丹参脂溶性多种丹参酮成分的高效液相测定方法。方法采用HP Hypersil ODS柱(250×4.0nm,5μm),丹参酮ⅡA洗脱流动相为甲醇-水(75∶25),二氢丹参酮I、隐丹参酮和丹参酮I为乙腈-水(50∶50),流速0.8ml/min,柱温控制37°C,丹参酮IIA检测波长为280nm,二氢丹参酮I、隐丹参酮和丹参酮I为254nm。结果四种丹参酮在各自浓度范围内线性关系良好(r>0.999);方法精密度、重复性良好;平均加样回收率大于98%,RSD小于3.02%。该方法下,样品在4h内稳定。三批精制冠心片的测定结果显示四种丹参酮成分的含量相对稳定。结论方法具有灵敏度高、可靠、准确和简便的特点,可用于精制冠心片中丹参脂溶性成分的测定。Objective To develop a high performance liquid chromatography （HPLC） method for quantitative determination of alcohol-soluble tashinones of Radix Salviae Miltiorrhizae in Jingzhi Guanxin Tablet. Methods The chromatographic analysis was performed on a HP Hypersil ODS column （250 × 4. 0 nm,5 μm） by elution with methanol-water（ 75 ： 25 ） for Tanshinone IIA and acetonitrile-water（50 ： 50） for Dihydrotanshinone I,Cryptotanshinone and Tanshinone I. The flow rate was 0. 8 mL/min, and the column temperature was maintained at 37 ℃. Detection was conducted at 280 nm for Tanshinone IIA and 254 nm for Dihydrotanshinone I, Cryptotanshinone and Tanshinone I. Results Good linear relationships were showed with correlation coefficients for the analytes exceeded 0. 999. Intra-assay and inter-assay precision of the analytes were less than 2. 96% ,and the average recovery rates obtained were more than 98% for all with RSDs below 3.02%. Quantitative analysis of three batches of Jinzhi Guanxin Tablet showed that the contents of the four tashinones were relatively consistent. Conclusion This well-developed HPLC method is reliable, reproducible and accurate, and can be used for quality control of Jinzhi Guanxin Tablet.
