成年豚鼠膀胱ICCs细胞起搏电流的鉴定

摘　　要：目的确定成年豚鼠膀胱卡哈尔间质细胞(interstitial cells of Cajal,ICCs)细胞是否具有起搏电流。方法采用胶原酶消化法,得到原代培养的成年豚鼠膀胱ICCs细胞。采用膜片钳技术,在全细胞记录模式下,记录其超级化激活的内向电流(hyperrepolarization-activated inward current,Ih)。结果原代培养的ICCs细胞和逼尿肌细胞的静息膜电位、膜电容存在明显差异;ICCs细胞可记录到起搏细胞特征电流Ih,而在逼尿肌细胞则记录不到。结论记录到成年豚鼠膀胱ICCs细胞具有起搏细胞的特征电流Ih,提示ICCs细胞在膀胱具有起搏作用。Objective To ascertain the presence of pacemaker-like current in interstitial cells of Cajal （ICCs） in the bladder of adult guinea pigs. Methods Primarily cultured bladder ICC of adult guinea pigs were harvested by collagenase digestion. Hyperrepolarization-activated inward current （Ih） and pacemaker specific current were recorded using whole-cell patch clamp technique. Results There was significant difference in resting membrane potential and membrane capacitance between ICCs and detrusor myocytes. Ih was confirmed in the ICCs, but not in the detrusor myocytes. Conclusion We have confirmed the existence of the specific pacemaker current in the ICCs in the bladder of adult guinea pigs, implying that ICCs might play an important role as pacemaker in the bladder.
