摘  要:在酸性条件下,大观霉素(SPM)与酸性铬蓝K(ACBK)、依文思蓝(EVB)及滂胺天蓝(POB)反应生成具有负吸收的红色或蓝色离子缔合物,最大褪色波长分别位于522、608和626nm,线性范围为0.2~5.0mg/L(ACBK)和0.1~5.4mg/L (EVB或POB体系),表观摩尔吸光系数(ε)分别为1.45×10^4、1.70×10^4和6.00×10^3L/(mol·cm);在一定浓度范嗣内,大观霉素的浓度与该几种体系的褪色度遵从朗伯比尔定律,据此建立了测定大观霉素的分光光度法;探讨了适宜的反应条件、方法的精密度、可靠性及其应用;方法用于大观霉素药物中大观霉素含量的测定,回收率分别为100%、98.8%和99.3%。Under acidic condition, spectinomycin (SPM) was reacted with acid chrome blue K (ACBK), evans blue(EVB) or pontamine sky blue (POB) to form red or blue ion association complexes showing obvious fading peaks. The maximum fading wavelengths were at 522, 608 and 626 nm, respectively, and the linear ranges were 0. 2 -5.0 mg/L(ACBK) and 0. 1 -5.4 mg/L(EVB or POB system), and their apparent molar absorptivities(ε) were 1.45 × 10^4 L/(mol · cm), 1.70 ×10^4 L/(mol · cm) and 6.00 × 10^3 L/(mol · cm), respectively. Under certain concentrations, the correlation of the spectinomycin concentration with the fading degree was found to obeyed Lombard Beer's law, and a new spectrophotometric method for the determination of spectinomycin(SPM) was thus developed. The optimum reaction conditions, the precision and reliability of methed and their applications were studied. The method was used for the determination of spectinomycin in injection samples and the recoveries of them were 100%, 98.8% and 99.3%, respectively.
