摘  要:目的观察丹酚酸B对大鼠局灶性脑缺血再灌注时神经细胞凋亡的影响及其可能的分子机制。方法健康雄性Wistar大鼠39只,随机分为3组,即假手术组、模型组和丹酚酸B组;线栓法制备大脑中动脉缺血再灌注模型;分别采用原位细胞凋亡检测技术和免疫组化方法,利用病理图像分析系统测定脑组织细胞凋亡指数(AI)以及bcl-2和bax蛋白表达量,应用半定量逆转录-聚合酶链反应(RT-PCR)检测各组大鼠脑组织bcl-2和bax mRNA相对表达量。结果丹酚酸B组AI较模型组降低,bcl-2表达增加,bax表达减少。结论丹酚酸B能上调bcl-2表达同时下调bax表达,这可能是其抗神经细胞凋亡的作用机制之一。Objective To study the effect of salvianolic acid B on neurocyte apoptosis and its possible molecule mechanism in rats with focal cerebral ischemia reperfusion injury, Methods Thirty-nine healthy male Wistar rats were randomly divided into 3 groups: sham-operation group, model group and sallvianolic acid B group, The intraluminal suture technique was used to establish the models of middle cerebral artery occlusion (MCAO) in rats. TUNEL method was used to detect apoptotic indexes (AI). The immunohistochemistry and pathological image analytical technique were used to moni- tor the expression of bcl-2 and bax proteins, and RT-PCR technique was used to monitor the expression of bcl-2 and bax mRNA in brain tissue of all groups of rats, Results In salvianolic acid B group, compared with model group, AI was decreased, and the expression of bcl-2 protein and mRNA was increased, while the expression of bax protein and mRNA was reduced. Conclusions Salvianolic acid B could up-regulate the expression of bcl-2 protein and mRNA, down-regu- late the expression of bax protein and mRNA simultaneously, which might be one of the mechanisms that salvianolic acid B resists neurocyte apoptosis.
