摘  要:目的:比较纳米级PHBV与PHBV/ECOFLEX复合支架材料在体外培养实验中对软骨细胞的影响,从而评价其作为软骨组织工程支架材料的可行性。方法:在纳米级PHBV聚集体与PHBV/ECOFLEX复合物上分别接种羊耳郭软骨细胞,并进行体外培养,取4h、24h、2d、4d、7d标本进行扫描电镜和组织学观察,评价软骨细胞的生长状况。结果:电镜及组织学观察均显示,纳米级PHBV支架材料中软骨细胞脱落较少,软骨细胞在支架表面黏附、铺展,并分泌细胞外基质;而PHBV/ECOFLEX复合材料塌陷明显,软骨细胞脱落较多,软骨细胞在支架表面未铺展。但2种支架材料内部的软骨细胞均较少。结论:纳米化处理的PHBV对软骨细胞有明显的吸附、诱导作用,可作为组织工程中的软骨修复支架材料。PURPOSE: To compare the potential use of nano-structured PHBV and PHBV/ECOFLEX composite as tissue engineering scaffold materials for chondrocyte regeneration. METHODS: Chondrocytes isolated from auricular cartilage of goat were seeded onto the surfaces of nano-structured PHBV and PHBV/ECOFLEX composite scaffolds. After culturing in vitro for 4 hours, 24 hours, 2 days, 4 days and 7 days, the chondrocytes-PHBV constructs were respectively examined histologically and under scanning electron microscope. RESULTS: The adhesion, growth and formation of extrocellular matrix on nano-structured PHBV were significantly better than those on PHBV/ECOFLEX composite. Chondrocytes inside the nano-structured PHBV were not found. CONCLUSIONS: Nano-structured PHBV is much better than PHBV/ECOFLEX composite as scaffold in cartilage tissue engineering in vitro system.
