摘 要:目的:探讨吡咯里西啶生物碱Jacoline对人正常肝L-02细胞内谷胱甘肽含量及谷胱甘肽还原酶(Glutathione Reductase,GR)活性的影响。方法:①采用DTNB法检测100μmol/L Jacoline与L-02细胞孵育24、48、72h后以及不同浓度Jacoline与L-02细胞孵育72h后对L-02细胞内谷胱甘肽、还原型谷胱甘肽(GSH)、氧化型谷胱甘肽(GSSG)含量及其GSH/GSSG的影响。②采用NADPH法检测100μmol/LJacoline作用24、48、72h后对L-02细胞内GR活性的影响。结果:①DTNB比色法显示,100μmol/LJacoline与L-02细胞孵育不同时间,谷胱甘肽和GSH含量随着时间的增加而减少,GSH/GSSG的比例也随时间的延长而减少;不同浓度的Jacoline与L-02细胞作用72h后,谷胱甘肽和GSH含量以及GSH/GSSG随浓度的增加而减少;②100μmol/L Jacoline与L-02孵育不同时间,GR的活性随着时间的增加而升高。结论:Jacoline对L-02细胞内谷胱甘肽和GSH含量、GSH/GSSG以及GR还原酶的活性有明显影响。Objective:To study the effects of pyrrolizidine alkaloid Jacoline on the level of cellular glutathione and the activity of glutathione reductase(GR) in human L-02 hepatocytes. Methods:The human L-02 hepatocytes were incubated with Jacoline 100 μmol/L, then the contents of glutathione, reduced glutathione(GSH), oxidized glutathione(GSSG) and GSH/GSSG were detected with 5, 5'-dithiobis-2- nitrobenzoic acid(DTNB) assay and the activity of GR was detected with NADPH assay at the 24, 48, 72 h respectively. The L-02 hepatocytes were also incubated with different concentrations of Jacoline for 72 h to detect the contents of glutathione, GSH, GSSG and GSH/GSSG. Results: With the time of incubation prolonged, the level of glutathione and GSH and the GSH/GSSG decreased, while the activity of GR increased. In comparison with different concentrations of Jacoline, after 72 h of incubation, the level of glutathione and GSH and the GSH/GSSG decreased with the concentration of Jacoline increased. Conclusion:Jacoline has significant effects on cellular glutathione, GSH, GSH/GSSG and the activity of GR in L-02 hepatocytes.
