摘 要:目的探讨益母草生物碱(Alkaloid of Leonurus,AFL)对去甲肾上腺素(norepinephrine,NE)诱导的新生大鼠心肌细胞肥大的作用。方法改良Simpson法培养新生大鼠心肌细胞,随机分为生理盐水对照组、NE组及AFL高中低剂量(0.05,0.1,0.5g/L)组。用NE诱导制作心肌肥大模型并给予相应的药物干预。采用伊红染色法测定心肌细胞表面积,BCA法测细胞蛋白含量,HOE33258荧光法测定细胞DNA含量,观察不同浓度AFL对肥大心肌细胞的影响。结果模型组细胞表面积、蛋白质含量及Protein/DNA比值与对照组相比明显增加(P<0.05);AFL中、高剂量组与模型组比较,细胞表面积、蛋白质含量及Protein/DNA比值均显著减少(P<0.05);低剂量AFL对细胞表面积、蛋白质含量及Protein/DNA比值无明显影响(P>0.05)。结论AFL可浓度依赖性地拮抗NE诱导的大鼠心肌细胞的肥大反应。Objective To observe the inhibitive effects of Alkaloid of Leonurus (AFL)on cardiaomyocyte hypertrophy induced by norepinephrine. Methods Cultured neonatal rat cardiomycytes were incubated with NE, AFL as the concentration of 0.05 to 0.5 g/L. Rat cardiomyocyte hypertrophic response were induced by NE and the anti- hypertrophic effects of AFL were assayed by measuring cell area, cell DNA content and protein content. The cells DNA content were measured by using HOE 33258 as a fluorescent indicator. Results NE significantly increased the cell area, protein content and Protein/DNA index in cultured cardiomyocyte (P〈O.05). AFL at 0.1 g/L and 0.Sg/L could significantly inhibit the cardiomyocyte hypertrophy induced by NE, decrease the cell area, protein content and Protein/DNA index (P〈 0.05). AFL at O.05g/L could not significantly decrease the cell area, protein content and Protein/DNA index (P〉0.05). Conclusion AFL could inhibit cardiomyocyte hypertrophyic response partly.
