摘  要:模拟河南省沿黄稻区水稻秸秆还田生态条件,分别在种植过水稻的潮土和没有种植过水稻的潮土中添加水稻秸秆培养70 d,在第0,5,25,45,70 d时采集土样.直接提取土样细菌总DNA,对细菌总DNA经PCR扩增后,对其产物进行DGGE分析.结果表明,施有水稻秸秆处理的土壤细菌群落多态性的变化远远复杂于空白对照土壤中的细菌群落变化,说明水稻秸秆还田能够增加土壤细菌群落分子多态性的丰富度;并且水稻秸秆刺激作用下不同土壤细菌群落分子多态性高峰出现时间不同.PCR and denaturing gradient gel electrophoresis (DGGE) were used to monitor the process of change and development of bacterial communities in paddy soil and aquic-fluvial soil, to which the rice straw or rice straw and microbiological agent were added during the incubating period, respectively. The total bacterial DNA from the two studied soils was extracted directly on 0, 5, 25, 45 and 70 d. The regions V3 of the 16S rDNA of bacteria from the two soils were amplified. DGGE profiles of the PCR productions were compared by similarity analysis. Rice straw was incubated in paddy soil and aquic-fluvial soil for 70 days. The results show that the composition of the bacterial community in soil with rice straw were more complex than CK during the incubating period. Meanwhile, the results showed that there were different pinks of the bacterial communities in the paddy soil and aquic-fluvial soil, to which rice straw were added during the incubation times.
