摘　　要：用抗人Ｂ细胞和Ｔ细胞共同分化抗原６Ａ８单克隆抗体从人扁桃体细胞λｇｔ１１ｃＤＮＡ文库克隆到１个长１３５８ｂｐ的ｃＤＮＡ。此ｃＤＮＡ内有一个长１２７８ｂｐ（２６～１３０３ｂｐ）的开放阅读框架，编码４２５个氨基酸的蛋白质。此蛋白质氨基酸序列中第１５７位到２２３位为疏水跨膜区。蛋白质的氨基酸序列与大鼠ＥＲα－甘露糖苷酶（１０４０个氨基酸）的６３２～１０３８位氨基酸序列间的相同性和相似性高达８２．０４５％和８８％。它与酵母Ｓａｃｃｈａｒｏｍｙｃｅｓｃｅｒｖｉｓｉａｅα－甘露糖苷酶（１０８３个氨基酸）的６５９～１０８３位氨基酸序列间的相同性和相似性也有３２．５％和５１．７５％。前者１６３～２７７位间与后者８３２～９４６位间１１５个氨基酸序列间的相同性和相似性达到５７％和７４％。由于６Ａ８ｃＤＮＡ核苷酸顺序未见于国外基因库资料，故已为ＮＩＨＧｅｎＢａｎｋ接受登记，接受号为Ｕ３７２４８。A 1358bp cDNA has been cloned from a human tonsil λgtll cDNA library by using a McAb 6A8 reactive with a differentiation antigen expressed on both human B and T cells.The cDNA contained an open reading frame from 26bp to 1303bp encoding a 425 amino acid protein with a hydrophobic region from 157 to 223 amino acid residue.The alignment showed an identity of 82% and a similarity of 88% between the amino acid(aa) sequences of the 6A8 cDNA encoded protein and the sequence from 632 1038 aa of a 1040 aa rat ER α mannosidase.The identity and similarity of the aa sequences between the 6A8 cDNA encoded protein and the sequence from 659 1083 of a 1083 aa yeast ( Saccharomyces cervisiae ) α mannosidase aa were 32 5% and 51 75%,respectively.Interestingly,the identity and the similarity between the 115 aa sequences from 163 to 277 aa of the 6A8 cDNA encoded protein and from 832 to 946 aa of the later reached to 57% and 74%.Since being not found in international gene banks,the 6A8 cDNA has been accessed by NIH GenBank with an accession number of U37248.
