小鼠胎肝基质细胞系MFLC克隆的建立、鉴定及细胞因子分泌

摘　　要：用体外长期培养的方法建立了小鼠胎肝基质细胞系ＭＦＬＣ，此细胞系是以上皮细胞为主的混合细胞系，能分泌多种细胞因子。为精确分析胎肝基质细胞在Ｔ细胞发育中的作用，对此细胞系进行了克隆化培养，共获得１１个克隆。对这些克隆进行角蛋白染色、倍增时间测定、染色体检查以及ＩＬ－６、趋化因子分泌水平的检测，结果表明，这些克隆在细胞学特性以及细胞因子的分泌上均有差异。A murine fetal liver stromal cell line,called MFLC,has been generated and maintained in DME supplemented with 10% NCS for 2 years.MFLC cell line is a transformed mixed stromal cell line in which the epithelial cells are dominant.The MFLC cells spontaneously secreted several types of cytokines,including IL 6,GM CSF and chemokine.Using the limited dilution method,the MFLC cell line was cloned,and 11 clones were derived.We have analyzed the characters and cytokine secretion of these clones.The results showed that these clones were different from one another in cell biological features and in the kinds and levels of cytokines secretion.
