摘  要:目的,建立用微量稀释法测定皮肤癣菌最低抑菌浓度(MIC)的方法学,为该类实验的标准化打下基础。方法:用96孔酶标板做药敏试验载体,10级倍比稀释药液,测定30株皮肤癣菌临床分离株对三唑类新药伊曲康唑和氟康唑的敏感性。结果:30株皮肤癣菌对伊曲康唑和氟康唑的MIC范围分别为≤0.06~4μg/ml和1~32μg/ml,均值分别为0.32μg/ml和7.76μg/ml.结论:用微量稀释法测定皮肤癣菌MIC初步显示了用药量少、重复性好、结果较可靠等优点。Objective: to set up a test methodology of determining MIC of dermatophytes by using a microdilution test, and provide a basis for the standardized tests in future. Methods: using 96 well microtiter trays as testing carriers, and ten twofold serial dilutions were made. The susceptibilities of 30 dermatophyte strains isolated from clinic to itraconazole and fluconazole, two new azole antifungal drugs, were determined. Results: the MIC range of itraconazole and fluconazole against 30 dermatophyte strains are 0.06~4μg/ml and 1~32μg/ml, respectively, with mean being 0.32μg/ml and 7.76μg/ml,respectively. Conclusion: the advantages of dermining dermatophyte's MIC to itraconazole and fluconazole by microdilution technique are economy of the test drugs and satifactory reproduction and neliability.
