摘 要:目的:观察莲房原花青素(LSPC)对人肝癌细胞HepG2生长及凋亡的作用。方法:以四甲基噻唑蓝来分析不同浓度LSPC对人肝癌细胞HepG2存活率的影响,用流式细胞术分析细胞周期和细胞凋亡,RT-PCR分析LSPC引起的生长抑制和DNA损伤基因GADD45β、GADD153、GADD34表达水平的变化。结果:LSPC可诱导HepG2细胞凋亡,抑制其生长,促进其凋亡(P<0.05),且呈浓度依赖性。生长抑制损伤基因GADD45β、GADD153、GADD34表达水平随着LSPC浓度增加而增加。结论:LSPC可抑制人肝癌细胞HepG2,具有抗肿瘤的药理作用。Objective To investigate the effects of lotus seedpol procyanidins (LSPC) on HepG2 cells growth and apoptosis. Methods HepG2 cells were exposed to different concentration of LSPC. Survival rate, cell cycle and cell apoptosis, expression level of growth-inhibition and DNA damage genes GADD45β, GADD153 and GADD34 were measured with ELISA, FCM and RT-PCR methods, respectively. Results The apoptosis of HepG2 cells was induced by LSPC. The cell growth was inhibited significantly by LSPC. In addition, these effects of LSPC were concentration-dependent. Expression levels of the three kinds of GADD genes increased with the concentration increase. Conclusions The results showed that LSPC can inhibit the growth of HepG2 cells and induce its apoptosis. It is suggested that LSPC has the ability of antitumor.
