摘 要:目的:研究智脑胶囊对血管性痴呆(VD)模型大鼠学习记忆及脑组织NO、ET、CGRP含量的影响。方法:反复夹闭双侧颈总动脉(CCA)结合腹腔注射硝普钠制备拟血管性痴呆大鼠模型,检测大鼠学习记忆能力及脑组织中NO、ET、CGRP含量。结果:与模型组相比,智脑胶囊3剂量组能明显改善实验性VD模型大鼠学习记忆能力;智脑胶囊中、高剂量组、脑复康组、尼莫地平组脑组织中NO含量明显减少;智脑胶囊3个剂量组、脑复康组、尼莫地平组均能够明显降低ET含量,增加CGRP含量。智脑胶囊高剂量降低ET和增加CGRP含量的作用明显优于脑复康,也明显优于智脑胶囊中、低剂量组。结论:智脑胶囊可改善实验性血管性痴呆大鼠学习记忆障碍,其机制与降低实验性VD模型大鼠脑组织中NO、ET含量,提高CGRP含量有关。Objective: To investigate the effect of Zhinao Capsule on behavior and the content of Nitric Oxide (NO), Endothelin (ET), Calcitonin gene related peptide (CGRP) in brain tissue of rats with vascular dementia. Methods: After intraperitoneal injection of low dose of sodium nitroprusside, animal models of vascular dementia were established by clamping common carotid arteries repeatedly. Their learning and memory ability and the content of NO, ET, CGRP of brain tissue were detected. Results: Compared with model group, three doses of Zhinao Capsule treated group could improve the ability of learning and memory obvioUsly; The middle and high dose of Zhinao Capsule treated group, piracetam treated group and nimodipine treated group could significantly decrease the content of NO; Zhinao Capsule treated group, piracetam treated group and nimodipine treated group could significantly decrease the content of ET and improve the content of CGRP. High dose of Zhinao Capsule could decrease the content of ET and the content of CGRP, which was more significant than the piracetam treated group and the middle and low dose of Zhinao Capsule treated group. Conclusions:Zhinao Capsule could improve the learning and memory ability in rats with vascular dementia, which may be related to decreasing the content of NO and ET and increasing the content of CGRP of brain tissue in rats with vascular dementia.
