摘  要:目的研究芫花根总黄酮(TFRD)抗肿瘤活性及其作用机制。方法以S180小鼠实体瘤对ICR小鼠造模。TFRD设3个剂量(25、50、75mg·kg-1),分别在造模的7d前、7d后和造模同时对小鼠灌胃14d。MTT法检测TFRD对体外培养的肿瘤细胞的毒性、给药后荷瘤小鼠脾淋巴细胞增殖和NK细胞的杀伤活性。结果3个剂量的TFRD、3种给药模式对小鼠S180肿瘤均的生长都表现出显著的抑制作用,对荷瘤小鼠的淋巴细胞增值、NK细胞的杀伤活性都有明显的提升作用,对体外培养的肿瘤细胞的细胞毒活性明显大于对正常细胞K293-T的细胞毒活性。结论TFRD具有显著的抗肿瘤活性,其抗肿瘤活性是通过对肿瘤细胞的选择毒性和提升机体免疫力实现的。Aim To study the antitumor activity and its possible mechanisms for action of total flavonoids from the roots of Daphne genkwa (TFRD). Methods ICR mice subcutaneously inoculated with S180 sarcoma were orally administered with three different doses of TFRD( 25,50 and75 mg·kg^-1·d^-1) for 14 days respectively in three different ways of treatment, i.e. the TFRD treatment was carried out 7 days before, during and after S180 sarcoma inoculation. MTT method was used for the assays of lymphocyte proliferation and the killing activity of NK cells. Results Three doses of TFRD all showed considerable inhibition to the growth of S180 sarcoma in three treatments, and significantly enhanced lymphocyte proliferation and the killing activity of NK cells in tumor-bearing mice. In addition, TFRD possessed higher cytotoxicity to tumor cell lines than K-293 cells. Conclusion TFRD is an agent with significant antitumor activity, and the mechanisms for its action are possibly mediated by the enhancement of lymphocyte proliferation and killing activity in tumor-bearing mice, as well as the selective toxicity to tumor cells .
