摘  要:目的探讨胸苷和茶碱在小鼠哮喘模型免疫调节中的作用。方法建立小鼠哮喘模型后,将动物随机分成三组,哮喘组、茶碱组及胸苷组。干预后第一、第二、第三周各处死1/3数量的动物。光镜观察其病理变化及支气管肺泡灌洗液(BALF)中细胞学分类;检测肺、脾组织中细胞凋亡情况;BALF上清液测定细胞因子IFN-γ、IL-4。结果与茶碱组相比,胸苷组早期肺部淋巴细胞(LC)和肺泡巨噬细胞(Am s)浸润明显,后期炎症明显恢复,且IFN-γ水平较其他两组均增高。结论胸苷能部分纠正Th2优势应答,对抗炎症反应。Aim: To discuss the immunore gulative effects of deoxythymldine and theophylline in mice models of asthma in order to provide a new clinical way to treat asthma. Methods 72 cases male Kunmln g mice were sensitized and challenged by ovabumin(OVA) as the mice models of asthmatic response; 1/3 mice(24 cases)were taken as the asthmatic response group mice,not treated; 1/3 mice were treated with theophylline, and the rest(24 cases)were treated with deoxythymidine. After they were treated for 7 days ,we executed 1/3 mice every week. Firstly, we explored the lung histopathological changes, detected the inflammatory cell apoptosis in lung and spleen with TUNEL technique,investigated the cytology changes and the level of relating cytokine (IFN-~, IL-4 )of bronchoalveolar lavage fluid (BALF). Results Compared with the theophylline mice,the histopathological changes of the deoxythymidine mice had a lot of L'C and AMs infiltrated in lung in early phase, whereas the inflammatory reaction in lung recovered significantly in last phase, and the level of IFN-~ in the group mice treated with deoxythymidine was significantly higher than that in the other two groups. Conclusion Deoxythymidine can decrease the inflammatory reaction in lung of the asthmatic mice.
