摘 要:目的:比较增生性瘢痕组织和正常皮肤组织刺激性G蛋白的表达差异,以明确刺激性G蛋白病理性瘢痕形成中的作用机制及其在哪些细胞中发挥作用。方法:实验于2005-09/2006-12在南昌大学第一附属医院烧伤研究所实验室完成。取10例18~38岁瘢痕整形手术患者术中剩余的全厚正常皮肤和切除的病理性瘢痕组织,采用PR—PCR法检测刺激性G蛋白的基因表达,免疫组织化学法检测G蛋白的表达,比较病理性瘢痕组织与正常皮肤组织间的差异。结果:①PR—PCR法检测发现两种组织均有刺激性G蛋白基因表达,利用凝胶图像处理系统分析,病理性瘢痕和内参照比值高于正常皮肤和内参照(P〈0.05)。②免疫组织化学检测后发现两种组织均有刺激性G蛋白的表达,均分布在表皮的基底细胞内、真皮的成纤维细胞内和毛细血管的内皮细胞内。在细胞内表达的部位符合G蛋白的本身性质(胞浆侧),两种组织的表达分布没有明显差异。③大体观察病理性瘢痕组织和正常皮肤的基底细胞刺激性G蛋白表达均为阳性,但无明显差异:真皮的成纤维细胞内和毛细血管内皮细胞内病理性瘢痕的表达强于正常皮肤。结论:刺激性G蛋白可能参与病理性瘢痕中成纤维细胞和毛细血管内皮细胞的增殖调控。AIM: To compare the expression difference in stimulatory G protein between hyperplastic scar and normal skin, so as to explore the formation mechanism of pathological scar. METHODS: The experiment was conducted in the laboratory of Burn Center, First Affiliated Hospital of Nanchang University from September 2005 to December 2006. All-thlckness normal skin and pathological scar were from patients aged 18-38 years old who underwent scar plastic operation. The expression of stimulatory G protein mRNA in sample was detected using PR-PCR method, and G protein expression was detected using immunohistochemical method. The expressions in pathological scar and normal skin were compared. RESULTS: ①Stimulatory G protein mRNA expression was found in both tissues by PR-PCR. But gel image processing system analysis showed that pathological scar Gs α / β -action was significantly higher than normal skin Gs α / β -action (P 〈 0.05). ②Immunohistochemical detection suggested that the expression of stimulatory G protein was found in both pathological scar and normal skin, distributing in epidermal basal cell, hypodermal fibroblast and blood capillary endothelial cell. The distribution of expression in the two tissues had no difference. ③In gross observation, positive expression in epidermal basal cell of pathological scar and normal skin had no obvious difference. The expression in hypodermal fibroblast and blood capillary endothelial cell of pathological scar was stronger than that in normal skin. CONCLUSION: Stimulatory G protein may play a role in the proliferation and regulation of fibroblast and blood capillary endothelial cell in pathological scar.
