摘  要:目的:有许多实验已证实,肌肉生长抑制素基因在骨骼肌中有特异性表达,在胚胎成体心脏以及乳腺组织中也检测到表达;但也有报道心脏、肺、脑、肾脏等多种组织中均未检测到肌肉生长抑制素的表达。本实验拟验证肌肉生长抑制素基因在大鼠不同组织中的表达情况。方法:实验于2006-11/2007-05在新乡医学院分子生物学实验室完成。①实验动物:2月龄清洁级SD大鼠,体质量240~280g。②实验方法:采用Trizol试剂从大鼠的肺脏、脑、骨骼肌和心脏组织中提取总RNA。③实验评估:采用半定量反转录-聚合酶链反应技术检测大鼠肺、脑、骨骼肌和心脏组织中肌肉生长抑制素基因的表达情况。结果:在肺、骨骼肌和心脏组织中均检测到肌肉生长抑制素的表达,以骨骼肌中的表达量最高,而在脑中未检测到。结论:肌肉生长抑制素在各个组织中表达情况不一,主要在骨骼肌中进行表达,是组织差异性表达基因。AIM: Many studies have shown that Myostatin gene is detected not only in skeletal muscle but also in the heart of adult and mammary gland in embryonic period. On the contrary, some studies have suggested that the Myostatin expression is not found in heart, lung, brain, kidney and other tissues. This paper investigated the expression of Myostatin gene in different tissues of rats. METHODS: The experiment was carded out in the Laboratory of Molecular Biology, Xinxiang Medical College from November 2006 to May 2007. The total Myostatin mRNA of lung, brain, skeletal muscle and heart tissue in 2-month-old SD rats of clean grade and 240 to 280 g were extracted using Trizol reagent and the expression was detected by semi-quantitative RT-PCR method. RESULTS: In the lung, heart and skeletal muscle tissues, the expressions of myostatin were detected, and the expression in skeletal muscle was the highest. No was detected in brain. CONCLUSION: The expression of Myostatin is different in various tissues. It is predominantly expressed in skeletal muscle, and it is a differentially expressed gene.
