摘 要:目的:观察血府逐瘀汤对气滞血瘀证模型大鼠血清蛋白质组表达的影响,初步探讨血府逐瘀汤在分子水平的作用机理。方法:复合因素制作大鼠气滞血瘀证模型,采集正常大鼠、气滞血瘀证大鼠及血府逐瘀汤治疗组大鼠的血清,应用一种简单的方法去除血清中的白蛋白,利用双向凝胶电泳分离血清总蛋白,通过Image Master 2D Elite 6.01凝胶图像分析软件分析2-DE图谱,比较各组间蛋白表达谱的改变情况。结果:正常组大鼠血清蛋白质点为(354±20),模型组大鼠血清的蛋白质点为(347±37),治疗组大鼠的蛋白质点数为(360±35),软件分析确定的差异蛋白点有7个,其中模型组大鼠血清与正常组相比有1个蛋白质下调,5个蛋白质上调,1个蛋白表达缺失,而给予血府逐瘀汤后可改善这些改变及缺失的蛋白质的表达水平。结论:血府逐瘀汤治疗气滞血瘀证的作用机制可能与这些差异表达的蛋白有关,这为血府逐瘀汤作用的分子机理研究提供了新的思路。Objective:To explore the impact of Xuefuzhuyv Decoction on serum proteome of rat with syndrome of Qi-stagnancy and blood stasis and to investigate the molecular mechanism of Xuefuzhuyv Decoction. Methods: The rat models with syndrome of Qi-stagnancy and blood stasis were established by combined methods. The rat serum was collected from normal group, model group and treated group. A simple method was employed to remove the high abundance albumin in serum. Proteomic protocol included proteome separation with two dimensional gel electrophoresis (2-DE), gel image analysis with Image Master 2D Elite 6.01 system, and identification of the different proteins between the three groups. Results: (354±20) proteins had been observed in normal group serum, (347±37) in model group serum, and (360±35) in treated group serum. Compared with normal group, seven changed proteins were determined: one protein decreased, five proteins increased and one protein lost in rat serum of model group, but these changes could be improved by treating Xuefuzhuyv Decoction. Conclusions:The molecular mechanism of Xuefuzhuyv Decoction which could treat the rat with syndrome of Qi-stagnancy and blood stasis may associated with those different proteins, and it will provide a new idea for study of molecular mechanism of Xuefuzhuyv Decoction.
