摘 要:目的:初步观察亚健康肾阴虚证患者血浆蛋白质组的变化与表达差异,为进一步探讨亚健康的发生机制奠定基础。方法:采用双向凝胶电泳(2-DE)技术分离亚健康肾阴虚证患者与正常人血浆总蛋白,银染显色;PDQuest软件对凝胶图象进行定性定量差异表达分析;从中选取差异表达蛋白质斑点,通过基质辅助激光解析电离飞行时间质谱(MALDL-TOF-MS)分析,获取肽质量指纹图谱(PMF);通过Mscot-Wizard软件进行数据库搜索,鉴定蛋白质。结果:获得了重复性和分辨性较好的血浆双向凝胶电泳图谱;在亚健康肾阴虚证血浆较正常人血浆表达量升高的蛋白质斑点11个,表达量降低的蛋白质斑点6个;选取其中表达量升高相差10倍以上的3个蛋白质斑点进行PMF鉴定,得到1个匹配精确的蛋白质(热休克蛋白27)。结论:亚健康肾阴虚证血浆与正常人血浆中存在差异表达蛋白质,利用蛋白质组学技术有望对亚健康的发生机制进行阐释,并将促进中医证候研究的拓展和深入。Objective: Observation sub-health deficiency Kidney-YIN of plasma proteomic expression changes with differences to further investigate the foundational mechanism of sub-health. Methods: Using two-dimensional gel electrophoresis (2-DE) separation technology sub-health deficiency in patints with normal plasma total protein, Silver stain color; PDQuest gel image software analysis for qualitative and quantitative differential expression; from the selection of differentially expressed protein spots, by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS), obtain peptied mass fingerpring (PMF); through Mscot- Wizard software database search to identification protein. Result: We obtained the satisfactory 2-DE patterns of sub-health deficiency Kindney-YIN. There axe 11 protein spots axe increased expression in Sub-health deficiency plasma than normal plasma, and 6 protein spots are reduced expression. Choose 3 proteome spots which expression increased more than 10 times to identification protein by PMF, obtained an exact match of protein (heat shock protein 27 ). Conclusion: Plasma sub-health deficiency with normal plasma protein ex- pression differences, The technology, of Proteomics is expected to be use to explain the foundational mechanism of sub-health. Pro- teomics will be the promotion of TCM research to the development and in-depth.
