摘 要:目的研究卵巢腺癌、黏液性和浆液性囊腺瘤及正常组织中IL-8mRNA,MCP-1mRNA和MIP-1αmRNA表达水平及其临床病理意义。方法43例卵巢腺癌、15例黏液性囊腺瘤、15例浆液性囊腺瘤和10例正常卵巢组织标本常规制作石蜡包埋切片,IL-8mRNA,MCP-1mRNA和MIP-1αmRNA染色方法均为原位杂交染色法。结果卵巢腺癌IL-8mRNA,MCP-1mRNA和MIP-1αmRNA表达阳性率明显高于黏液性或浆液性囊腺瘤及正常组织(P<0.05或P<0.01);组织学分级G1、临床分期Ⅰ+Ⅱ、无淋巴结转移及未侵犯周围组织器官的卵巢腺癌病例IL-8mRNA,MCP-1mRNA和MIP-1αmRNA表达阳性率明显低于组织学分级G3、临床分期Ⅲ+Ⅳ、淋巴结转移及侵犯周围组织器官病例(P<0.05或P<0.01);卵巢腺癌中IL-8mRNA,MCP-1mRNA和MIP-1αmRNAM之间呈高度一致性(P<0.01)。结论IL-8mRNA,MCP-1mRNA和MIP-1αmRNA表达可能是反映卵巢腺癌发生、进展、生物学行为及其预后的重要化学趋化因子。Objective To study the expressive levels of IL - 8mRNA, MCP - 1 mRNA and MIP - 1 mRNA and their clinicopathological significances in adenocarcinoma, mucions cystoadenoma, serous cystoadenoma and normal tissues of ovarian. Methods In situ hybridization was used for detecting expressive levels of IL - 8mRNA, MCP - 1 mRNA, MIP - lctmRNA in routinely paraffin - embedded sections from specimens of ovarian adenocarcinoma ( n = 43 ) , mutinous cystoadenoma ( n = 15 ) , serous cystoadenoma ( n = 15 ) , and normal tissues ( n = 10). Results The expressive rates of IL - 8mRNA, MCP - 1 mRNA and MIP - 1 αmRNA were significantly higher in ovarian adenocarcinoma than those in mutinous cystoadenoma and normal tissues (P 〈0.05 or P 〈0.01 ). The expressive postitive rates of IL- 8mRNA,MCP -1 mRNA and MIP -1 αmRNA were significantly lower in the cases of histologic grade G1, clinical stage Ⅰ + Ⅱ , no - metastasis of regional lymph node, and no - infiltration of peritumoral tissues than those in the ones of histologic grade G3 , clinical stage Ⅲ + Ⅳ, metastasis of regional lymph node, and infiltration of peritumoral tissues in ovarian adenocarcioma ( P 〈 0.05 or P 〈 0.01 ). The high consistances were found among the expressive levels of IL- 8mRNA,MCP -1 mRNA and MIP -1 αmRNA in ovarian adenocarcinoma tissues. Conclusion The exprssions of IL - 8mRNA, MCP - 1 mRNA and/or MIP - 1 αmRNA might be important chemokines for reflecting the carcinogenesis, progression, biological behaviors and prognosis of ovarian adenocarcinoma.
