摘  要:目的:探讨核因子κB受体活化剂配体RANKL在大鼠下颌第一磨牙牙胚冠方组织中mRNA的表达及破骨细胞的分化情况。方法:运用原位杂交法检测大鼠下颌第一磨牙牙胚冠方组织中RANKL mRNA的表达;TRAP染色观察破骨细胞分化情况。结果:出生1、、5、d大鼠下颌第一磨牙牙胚冠方牙囊成纤维细胞、成釉细胞RANKL mRNA37的阳性表达强于对照组牙龈成纤维细胞(p<0.01)。大鼠下颌第一磨牙牙胚冠方出生后1d破骨细胞多为单核,3d时多核破骨细胞增多。结论:RANKL mRNA在大鼠出生后1、、5、d下颌第一磨牙牙囊成纤维细胞、成釉细胞中有表达,37可能通过促进破骨细胞的分化及成熟参与牙齿的萌出。Objective:To observe the expression of the receptor activator of the nuclear factor-kappa B ligand(RANKL) mRNA at the occlusal surface of tooth germ in rat mandibular first molar and the differentiation of osteoclast. Method:RANKL mRNA was observed at the occlusal surface of tooth germ by in situ hybridization;The differentiation of osteoclast was observed by tartrate-resistant acid phosphatase in mouse mandibular first molar. Result:RANKL mRNA positive signals in the fisrt, third,fifth and seventh days were observed in tooth germ fibroblast and ameloblast monocyte in first day and multinucleated cell in the third day. Conclusion:RANKL mRNA of tooth germ fibroblast and ameloblast express positive signals in fisrt,third,fifth and seventh day. RANKL was likely to participate in tooth eruption throught differentiation and martue of osteoclast.
