牛大力多糖对小鼠T淋巴细胞增殖的双向调节作用

摘　　要：目的从牛大力(Millettia Speciosa Champ.)提取纯化多糖,流式细胞术检测多糖对刀豆蛋白A(Con A)刺激引起的小鼠T淋巴细胞增殖的影响,探讨牛大力多糖对免疫系统的调节作用机制,为临床用牛大力治疗多种慢性疾病提供理论和实验依据。方法提取、纯化牛大力多糖,利用CFDA-SE染色,流式细胞术检测多糖对Con A刺激引起的小鼠T淋巴细胞增殖的影响。结果终浓度为160μg/mL、800μg/mL、4 mg/mL、20 mg/mL的多糖对小鼠T淋巴细胞增殖呈剂量依赖性抑制,而终浓度为5、25、50、125μg/mL的多糖对小鼠T淋巴细胞增殖起促进作用,剂量依赖关系不明显。结论牛大力多糖对小鼠T淋巴细胞的增殖呈双向调节作用。Objective The polysaccharide was extracted and purified from MiUettia Speciosa Champ. Flow cytometry was used to investigated the effects of polysaccharide on mouse lymphocyte proliferation stimulated by Con A, the mechanism of the effects of polysaccharide on the immune system was explored and theoretical and experimental evidence for the clinical application of MiUettia Speciosa Champ in treating chronic diseases were drawn. Methods The polysaccharide was extracted and purified from MiUettia Speciosa Champ,CFDA-SE staining and flow cytometry were used to detect the effects of polysaccharide on mouse lymphocyte proliferation stimulated by Con A, Results The polysaccharide has the function to inhibit the proliferation of lymphocyte of mouse depending on its concentration with 160μg/mL,800μg/mL,4 mg/mL,20 mg/mL in substrate, but promotes the proliferation of T lymphocytes of mice lymph node at the concentrations of 5,25,50, and 125μg/mL,, but the dose-dependent effect is not significant. Conclusion The polysacchafide extracted from MiUettia Speciosa Champ has the two-way effects on T lymphocyte proliferation in mice lymph node stimulated by Con A.
