淫羊藿苷对高脂血症大鼠粘附分子基因表达的影响

摘　　要：目的研究淫羊藿苷对高脂血症大鼠粘附分子基因表达的影响。方法健康雄性SD大鼠40只,随机分为对照组、高脂饮食组、高脂饮食+低剂量淫羊藿苷组、高脂饮食+高剂量淫羊藿苷组,检测各组大鼠血清脂质水平及观察主动脉病理学改变。并采用荧光实时聚合酶链反应法比较主动脉壁细胞间粘附分子1和血管细胞粘附分子1基因的表达。结果与高脂饮食组比较,淫羊藿苷低剂量组和高剂量组总胆固醇、甘油三酯和低密度脂蛋白胆固醇水平(P<0.05或0.01)降低,细胞间粘附分子1mRNA的表达显著减低(P<0.01),而血管细胞粘附分子1mRNA的表达没有显著改变(P>0.05)。结论淫羊藿苷能降低高脂血症大鼠血脂并抑制细胞间粘附分子1mRNA的表达。Aim To investigate the influence of ieariin on adhesion molecule gene expression in hypedipidemia rats, Methods 40 male healthy SD rats were randomly divided into four groups： control group, high cholesterol group, high cholesterol with low dose icariin （ 100 mg/kg）group, high cholesterol with high dose icariin （200 mg/kg）group. Serum lipid level of all rats was measured and the aorta was taken for pathologic analysis. The mRNA expression of both intercellular adhesion molecule- 1 （ICAM-1） and vascular cell adhesion molecule-1 （VCAM-1） on the aorta was analyzed by realtimr PCR. Results As compared with the high cholesterol group, the level of total cholesterol （TC）, triglycefide （TG） and low density lipaprotein cholesterol （ LDLC） in high cholesterol with icariin was obviously lower （ P 〈 0.05 or 0.01 ） ; the mRNA expression of ICAM- 1 was significantly decreased （P 〈 0.01 ） ; but the mRNA expression of VCAM- 1 was not significantly inhibited. Conclusion Icariin can reduce the level of blood lipid and the mRNA expression of ICAM-1 in hyperlipidemia rats.
