摘 要:为了解猪瘟病毒(CSFV)石门株强毒感染对免疫细胞的影响以及免疫细胞的变化动力学,用流式细胞仪对人工感染猪的白细胞凋亡以及CD4+和CD8+T淋巴细胞亚群的动态变化规律进行了检测。结果表明,攻毒后CD4+和CD8+T淋巴细胞比例急剧下降,CD4+T淋巴细胞比例在攻毒后第4 d和第7 d分别由攻毒前的26.87%下降到8.83%和13.86%,CD8+T淋巴细胞比例由攻毒前的28.17%分别下降到14.19%和17.55%。外周血白细胞凋亡的检测结果显示,发病初期感染组凋亡细胞和早期凋亡细胞均显著高于对照组,表明CSFV诱导细胞凋亡是导致免疫细胞减少和免疫抑制的主要原因。To elucidate the effects of virulent classical swine fever virus (CSFV) Shimen strain infection on immune cells and to understand their kinetics,kinetics of CD4^+ and CD8^+ T lymphocyte subpopulations and peripheral blood leukocytes apoptosis of pigs infected experimentally with CSFV were analyzed using flow cytometry. The results showed that CD4^+ and CD8^+T lymphocyte subpopulations were significantly reduced,the percentage of CD4^+T lymphocyte subpopulation decreased from 26.87% prior to infection to 8.83% on day 4 post-infection(PI) and 13.86% on day 7 PI,respectively,whereas CD8^+ T lymphocyte subpopulation from 28.17% prior to infection to 14.19% on day 4 PI and 17.55% on day 7 PI,respectively. Counts of apoptotic leukocytes and early apoptotic leukocytes in peripheral blood of the infected pigs were significantly more than those of the control pigs,indicating that apoptosis induced by CSFV infection was the major cause of leucopenia and immune suppression.
