大鼠急性肾功能衰竭中细胞凋亡的实验观察及其意义

摘　　要：目的:观察在甘油致急性肾功能衰竭大鼠中,肾小管上皮细胞凋亡的变化与发生途径。方法:采用肌注甘油法建立急性肾功能衰竭大鼠模型,分24h与5d两个时点取材检测血浆尿素氮(BUN)、血肌酐(Scr)、肾组织脂质过氧化物(LPO),并进行肾组织凋亡细胞流式细胞术检测以及彗星实验。结果:在24h时点,急性肾功能衰竭大鼠BUN、Scr水平均明显升高,彗星实验阳性细胞较正常水平明显升高,第5天,BUN、Scr、肾组织LPO水平基本趋于正常,但肾小管上皮细胞凋亡持续存在,彗星实验阳性细胞依然有维持较高水平的趋势。结论:急性肾衰的损伤功能障碍与修复各阶段细胞凋亡充当不同的角色,其诱导细胞凋亡的机制也可能不同。Objective：To investigate the change of apoptosis of the rats with acute renal failure. Methods： The model of acute renal failure was established by injecting glycerol into the muscle of Wistar rats. BUN and Scr of plasma, LPO, DNA damage and apoptosis of the renal tissue were assayed by automatic biochemistry analysis, TBA chromatometry, Comet assay, and FCM. These indexes were detected at 24 h and 5 d after acute renal injury. Result：The levels of BUN, Scr, DNA damage and LPO were increased at 24 h, and recovered at 5 d. Apoptosis of the renal tissue expressed highly in the different stages of acute renal failure. Conclusion：In different stages of acute renal failure, apoptosis can play the different role.
