摘 要:目的:了解异种冻干脱钙骨基质(xenogeneic demineralized freeze-dried bone matrix XDBM)与骨膜成骨细胞复合移植的新骨形成能力。方法:将XDBM与成骨细胞复合后的组织工程骨和单纯XDBM分别植入24只新西兰兔双侧后腿肌袋内,术后2、4、6、8周取植入材料作组织学和骨矿物含量测定。结果:XDBM与成骨细胞复合体植入后2周有软骨和新生骨形成,4周时发育成编织骨并形成髓腔,术后6、8周,新生骨进一步成熟、骨量增多;移植物钙含量随时间推移逐渐增高。整个观察过程,单纯植入XDBM组肌袋内无新生骨或软骨形成,移植物被纤维组织包裹?浸润,钙含量持续处于较低水平。结论:XDBM单独移植无异位成骨能力;XDBM与成骨细胞复合后具有很好的异位成骨能力,XDBM是组织工程骨研究较理想的支架材料。Objective:To investigate the abilities of ectopic osteogenesis by periosteal osteoblasts combined with xenogeneic demlneralized freeze-dried bone matrix(XDBM). Methods. Tissue engineered bones by osteoblasts co-cultured with XDBM were implanted into the bilateral hind limb muscle pocket of New Zealand rabbits, XDBM implanted alone was used as control. The implanted materials were taken out at 2,4,6,8 weeks postoperation. Histological and calcium contents were assessed. Result: Cartilage mixed new bone tissues apperienced in tissue engineered bone implantation group at 2 weeks, the grafts turned into woven bone and medullary cavities appeared at 4 weeks, the quantities of new bones formation increased in 6-8 weeks postoperation, calcium contents of the implantation increased as time went on in this group. For control group of XDBM implantation alone, the implanted materials were filled with fibrous tissue, and no new bones or cartilages can be found. Calcium contents of the implanted materials remained low level through the observed duration. Conclusion: XDBM cannot induce of ectopic bone formation. Supplementation of XDBM with osteoblasts significantly increased abilities of ectopic osteogenesis. XDBM can be used as extracellular scaffold in bone tissue engineering.
