摘  要:目的观察粉防己碱对培养的人增生性瘢痕成纤维细胞增殖及周期的影响。方法培养人体增生性瘢痕的成纤维细胞,设实验组和对照组,实验组加入粉防己碱,用四甲基偶氮唑盐(MTT)法观察细胞的增殖率,用流式细胞仪观察细胞周期,比较两组之间的细胞增殖率和细胞周期。结果细胞增殖率实验组吸光度为(0.31±0.03),对照组为(0.53±0.04),两组比较差异有统计学意义(P<0.05);对照组处于增殖期的细胞占细胞总数的68.3%,实验组处于增殖期的细胞明显减少,为占总数的40.8%,两组比较差异有统计学意义(P<0.05)。结论粉防己碱通过阻止成纤维细胞由静止期进入增殖期而抑制其增殖,可能具有防治瘢痕的作用。Purpose To investigate the effects of Tetrandrine on proliferation and cycle of human hypertrophic scar-derived fibrobiasts. Methods Hypertrophic scar-derived fibroblasts were harvested from the patients and cultured in vitro. Tetrandrine was added to the culture media in the experimental group. The rates of cell proliferation and the cell cycles were compared between the experimental and the control groups using MMT and FCM after 24 hours of culture. Results The rates of cell proliferation ( optical density )were significantly decreased after the use of Tetrandrine(0.31 ± 0.03)for the experimental group versus(0.53±0.04)for the control group, P 〈 0.05) and the cell number of proliferative phase in the experimental group was significantly reduced than in the control group(40.8 % versus 68.3 %, P 〈 0.05 ). Conclusion Tetrandrine can inhabit proliferation of human hypertrophic scar-derived fibroblasts by restraining them from entering proliferative phase, and it might prevent and treat the sears.
