五味地龙汤对实验性哮喘豚鼠血清干扰素-γ水平的影响

摘　　要：目的:探讨五味地龙汤对哮喘的作用及其机制。方法:按豚鼠γ-干扰素(IFN-γ)ELISA试剂盒说明,采用双抗夹心ELISA法,测定血清IFN-γ含量。结果:豚鼠鸡卵清蛋白致敏并激发后出现活动减少,反应迟钝,毛色失去光泽,呼吸次数增加,呼吸困难。药物组各组均无明显变化。五味地龙汤大剂量组豚鼠血清IFN-γ为(35.06±2.83)pg.ml-1,五味地龙汤中剂量组血清IFN-γ为(29.74±1.84)pg.ml-1,五味地龙汤小剂量组血清IFN-γ为(28.80±1.36)pg.ml-1,地塞米松组血清IFN-γ为(39.08±1.06)pg.ml-1,与哮喘模型组血清IFN-γ(18.74±2.22)pg.ml-1比较,差异均有统计学意义(P<0.01)。五味地龙汤小剂量组与模型组比较,血清IFN-γ差异有统计学意义(P<0.05)。结论:五味地龙汤能升高哮喘动物血清IFN-γ水平,其作用与剂量成正相关。这可能是五味地龙汤间接抑制炎性介质的释放,从而达到治疗气道炎症的机制。Objective: To explore effect and mechanisms of Schisandra fluid and Earthworm decoction on asthma.Methods: The taken intracarotid blood was centrifugated at velocity at 3000r·min-1 for 10 min, from which the serum was taken.The level of serum interferon-γ(IFN-γ)was determined by double antibody sandwich ELISA way with the guinea pig IFN-γkit.Results: The model guinea pigs repeated with OVA intranasal challenge moved about decreasing,reaction retardation,lustre of coat of hair lost,tachypnea and dyspneic respiration,but another guinea pigs were a good state.Serum IFN-γin Schisandra fluid and Earthworm decoction large dosage group(35.06±2.83) pg·ml-1,middle dosage group(29.74±1.84) pg·ml-1,small dosage group(28.80±1.36) pg·ml-1 and in dexamethasone group(39.08±1.06) pg·ml-1 were very significantly raised as compared to that in model group(18.74±2.22) pg·ml-1(all P<0.01).It was a significant difference between the Schisandra fluid and Earthworm decoction small dosage group and large dosage group(P<0.05),too.Conclusion: It was demonstrated that Schisandra fluid and Earthworm decoction can heighten the level of serum IFN-γin asthma guinea pigs.It was that the decoction can inhibit directly and indirectly inflammatory media to release and cure the airway inflammation.The efficiency is a direct proportion.
