摘 要:目的观察膈下逐瘀汤对肝硬化大鼠一氧化氮(NO)、内皮素-1(ET-1)及血栓素B2(TXB2)、6-酮-前列腺素F1α(6-Keto-PGF1α)含量的影响。方法将50只SD大鼠随机分为5组,正常对照组、模型对照组、膈下逐瘀汤高剂量组、膈下逐瘀汤低剂量组、复方鳖甲软肝片组各10只。除正常对照组外,其余各组采用四氯化碳(CCl4)、普通饮食及乙醇建立大鼠肝硬化模型。造模成功后,各组分别予相应药物灌胃,正常对照组、模型对照组予0.9%氯化钠注射液,灌胃容积均为1mL/只,每日给药1次,连续灌胃4周。检测各组ET-1、NO、TXB2、6-Keto-PGF1α的含量,并进行统计分析。结果膈下逐瘀汤高剂量组ET-1、NO、TXB2、6-Keto-PGF1α明显降低,与模型对照组、复方鳖甲软肝片组、膈下逐瘀汤低剂量组比较差异均有统计学意义(P<0.05,P<0.01)。结论膈下逐瘀汤可通过有效地抑制NO、ET-1的产生,恢复血管平滑肌的正常舒缩状态,改善肝脏血流动力学;可显著降低肝硬化大鼠血清TXB2和6-Keto-PGF1α含量,调节二者的比例,使其达到适宜的有效浓度,以消除血小板聚集和血栓形成,改善肝脏微循环。Objective To observe the effect of Gexiazhuyu decoction on the levels of NO, ET- 1, TXB2 and 6-Keto-PGF1α in rats with hepatic cirrhosis. Methods The rat models with hepatocirrhosis were established with seretin, routine diet and alcohol. After the models were successfully set up, the rats were divided into normal control group, model control group, high- dose Gexiazhuyu decoction group, low- dose Gexiazhuyu decoction group and compound Biejiaruangan tablets group. The levels of ET - 1, NO, TXB2, 6-Keto-PGF1α were detected and statistics analysis was conducted. Resuits The levels of ET - 1, NO, TXB2, 6-Keto-PGF1α in high - dose Gexiazhuyu decoction group was significantly lower than those in other three groups ( P 〈 0. 05 or P 〈 0. 01 ). Conclusion Gexiazhuyu decoction can recover the nomud diastolic and contractile state of vascular smooth muscle, and improve the hemodynamic of liver by inhibiting effectively the generation of No and ET - 1. Gexiazhuyu decoction can reduce considerably the content of serum TXB2 and 6-Keto-PGF1α ; regulate the ratio of the two factors to suitable effective concentration, in order to eliminate the aggregation of blood platelet and thrombosis as well as improve micro circulation of liver.
