摘  要:目的建立及评价喂养型肥胖相关肾病(ORG)大鼠动物模型。方法将30只雄性Wistar大鼠随机分为2组:正常对照组(n=10)和模型组(n=20)。正常对照组用普通饲料,模型组用高脂饲料各喂养24周,观察大鼠体重及Lee’s指数、血压、血糖、血脂、24h尿蛋白定量、血胰岛素、瘦素及肾脏病理改变。结果造模24周末,模型组体重、Lee’s指数显著高于正常对照组(P<0.01),尾动脉血压、24h尿蛋白定量、空腹血胰岛素及瘦素均明显高于正常对照组(P<0.01),三酰甘油及血浆游离脂肪酸亦高于正常对照组(P<0.05)。而两组间总胆固醇在整个饲养期均无统计学意义差异(P>0.05),两组大鼠空腹血糖均在正常范围内,但模型组大鼠空腹血糖亦高于正常对照组(P<0.05)。模型组大鼠肾脏病理变化明显,肾小球囊腔增大(P<0.01),基底膜增厚(P<0.01)。结论各项指标及肾脏病理证明,用高脂饲料可以制备出符合或接近临床的ORG动物模型。Objective To establish and evaluate a new rat model of obesity related glomerulopathy(ORG). Methods Thirty male Wistar rats were randomly divided into two groups: normal control group( NC, n = 10)and model group(ORG, n = 20 ). The rats in NC group and ORG group were fed with underlying forage and high lipid forage for 24 weeks, respectively. Body weight(BW), Lee' s index number, blood pressure (BP), fasting plasma glucose ( FBG ), total cholesterol ( TC ), triglyceride (TG), free fatty acids (FFA), 24 h urine albumen( 24 h UALb), fasting insulin(FINS) and leptin(LEP) of rats were measured. The renal pathological changes were observed at week 24 by optics microscope and eleetron microscope. Results Compared with NC group, body weight, Lee's index number, BP, 24 UALb,serum levels of FINS and LEP increased significantly in ORG group at week 24(P〈0.01) ,and serum levels of TG and FFA were higher( P 〈 0.05). TC had no significant difference between two groups( P 〉 0.05). FBG in two groups was within normal limits, but higher in ORG group than in NC group( P 〈 0.05). Renal pathology in ORG group showed a more tremendous change than those in NC group. In ORG group glomerular capsule space increased(P 〈 0.01) ,and glomerular base- ment membrane was a little thicker( P 〈 0.01 ). Conclusion The results suggest that the high lipid diet feeding could successfully establish the rat model of ORG for clinical practice.
