摘  要:目的:建立高效液相色谱-串联质谱分析方法,研究丹参-郁金中丹参素在兔体内的组织分布。方法:免灌胃给予丹参-郁金水煎液后,于30,50,70min处死,取心、脑、肝、肾、肺、脾组织,10%高氯酸沉淀组织蛋白,上清液2倍量乙酸乙酯萃取3次,氮气吹干,残渣用流动相溶解,高效液相色谱-串联质谱法测定各组织中的丹参素含量。结果:检测限为0.01μg/mL,线性范围为0.05~400.0μg/mL,方法回收率为83.2%~105.6%;丹参中丹参素在各组织中的分布特征为:肾〉脑〉肝〉脾〉肺〉心。结论:该方法精密、灵敏、准确,可用于其他生物样品中丹参素的测定。Aim: To establish a high performance liquid chromatography-trap mass spectrum (LC-MS/MS) method and to study tissue distribution of Dartshertsu in Huoxue Huayu formulae in rabbits. Methods: After intragastric administration of Salvia mitiorrhiza and Radix Curcumae decoction, rabbits, sacrificed by decapitation at 30, 50 and 70 min respectively, were exposed to rapid removal of heart, brain, liver, kidney, lung, and spleen. Homogenated tissues were pretreated by 10% perchloric acid, and the resulted supernatants were extracted three times using ethyl acetate with the volume two times of the supernatant. The ethyl acetate solution was evaporated to dryness under a flow of nitrogen. The residue was reconstituted in the mobile phase. LC-MS/MS was employed for the determination of Danshensu in heart, liver, kidney, spleen, lung and brain. Results: The limit of detection of Danshertsu in these samples was 0.01μg/mL, and the linear range was 0.05- 400.0 μg/mL. The method recoveries were found to be 83.2% -105.6%. The characteristic tissue distribution of Danshertsu in rabbits was presented as the decreasing order of kidney, brain, liver, spleen, lung and heart. Conclusion: It is a precise, sensitive and accurate method which could be used for the determination of Danshensu in biological samples.
