肠道上皮细胞损伤与Th1分泌干扰素γ关系的研究

摘  要:目的探讨辅助性T细胞在肠黏膜损伤中的作用。方法大鼠上皮细胞系(IEC-6)用刮匙轻刮IEC-6单细胞层产生"十"字缺损区,分别将大鼠重组IFN-γ(5ng/ml)和IL-4(5ng/ml)加入到细胞培养液中,培养IEC-6细胞16h,观察细胞缺损区的面积,评价不同因子对细胞损伤后修复的影响。结果IFN-γ抑制EC-6细胞损伤后的修复(P<0.05);在有或无H2O20.25mM氧化应激的情况下,IL-4可以促进IEC-6细胞损伤后的修复,但与对照组相比差异无统计学意义(P>0.05)。结论Th1细胞可能通过分泌INF-γ调节IEC-6细胞损伤后的修复。Objective To investigate the effects of Thl during intestinal epithelial injury. Methods A denuded epithelial "crosscenter" wound was created in a standardized fashion by scraping the IEC - 6 monolayers with a gel loading pipet tip. Rat epithelial cell line (IEC - 6 cells ) was treated with IFN -γ(5ng/ml) or IL - 4( 5ng/ml ). The denuded wound area (ROI) was quantified. Wound repair was calculated as the reciprocal of denuded wound ROI. Result IEC - 6 cells treated with IFN - γ inhibited wound repair( P 〈 0.05), in contrast, IL - 4 promoted IEC - 6 cell wound repair. Conclusions IFN -γ is involved in inhibition of epithelial repair in IEC - 6 cells. Thl regulates epithelial repair of IEC - 6 cells by secreting.
