摘 要:为了从药用植物葛的内生真菌中寻找天然抗氧化剂,在葛(Pueraria lobata)的根、茎、叶以及果实中分离出128株内生真菌,并采用DPPH(二苯基苦基苯肼)自由基酶标仪法对其清除自由基活性进行检测。结果表明,有多株内生真菌具有不同程度的清除自由基活性,且不同产地以及不同部位分离出的内生真菌活性不同;同种菌株清除自由基活性与提取液浓度、反应时间成正比关系,即样品浓度增加活性增强,反应时间增长其活性也增强。其中菌株10y-4在浓度2.5 mg.mL-1、反应时间为20 min时,其清除自由基活性达到92.3%。表明葛内生真菌可作为天然的高效抗氧化剂进行开发利用。We isolated 128 strains of endophytic fungus from the root, stem,leaf and fruit of Pueraria lobata to inspect it free radical scavenging activities by using DPPH-microplate assay and DPPH-TLC assay methods. The resuits showed that many strains of endophytic fungus have free radical scavenging activities, and the endophytic fun- gus isolated from different parts of Pueraria lobata and taken from different places have different activities. At the same time, the free radical scavenging activities of the strains are directly in proportion to the extract concentration and reaction time, i.e. the activity increases with the increase of the concentration and its reaction time. The free radical scavenging activity of strain 10y-4 can reaches 92. 3% after incubating for 20 rain at 2. 5 mg·L^-1 concentration. The research showed that endophytic fungus taken from Pueraria lobata could be developed and used for natural, high-efficient free radical scavenging agent.
