摘  要:[目的]研究壬基酚(NP)对性成熟期F_1雄性SD大鼠睾丸支持细胞功能的影响机制。[方法]设50、100、200mg/kg NP 3个剂量染毒组和1个对照组,对母鼠从妊娠第7天至断乳期灌胃染毒,产后55天处死雄性子代,取睾丸固定制片后,分别作病理组织学观察,作波形蛋白(vimentin)、广谱钙黏附蛋白(pan-cadherin)、信号转换和转录激活因子3(STAT3)的免疫组化分析和雄激素结合蛋白(ABP)、白细胞介素-6(IL-6)的原位杂交试验。[结果]睾丸组织病理学检查显示,各剂量染毒组发育异常的生精小管数增多,支持细胞数量减少;200mg/kg组波形蛋白、广谱钙黏附蛋白的表达低于对照组(P<0.05);各染毒组STAT3的表达与对照组相比差异无统计学意义;100mg/kg和200mg/kg组ABP mRNA和IL-6 mRNA的表达低于对照组(P<0.05)。[结论]200mg/kg NP可抑制性成熟期F_1雄性SD大鼠支持细胞的波形蛋白、广谱钙黏附蛋白、IL-6、ABP的表达,影响支持细胞功能。[ Objective ] To investigate the adverse effects and its mechanism of nonylphenol( NP )administered to mother SD rats on Sertoli Cells of their male offspring in sexual maturation period. [ Methods ] There were 4 dose groups including control, 50, 100 and 200mg/kg NP groups respectively. NP was administered to dams by gavage from gestation day 7 to weaning period. Pups were sacrificed at postnatal day 55. Testes were collected and histopathological examination performed. Then ABP mRNA and IL-6 mRNA levels were examined by tissue in situ hybridization. The expression of vimentin, pan-cadherin and STAT3 were examined by immunohistochemical analysis. [ Results ] Histopathology revealed that NP-treated groups had higher rates of maldeveloped seminiferous tubules, lower number of Sertoli cells than the control. Compared with control, vimentin and pancadherin expression were significantly decreased in 200mg/kg group( P 〈 0.05 )while there was no significant difference in STAT3 expression between the control and NP-treated groups. The level of ABP mRNA and IL-6 mRNA decreased in 100 and 200mg/kg groups( P 〈 0.05 )when compared with that in control. [ Conclusion ] 200mg/kg NP apparently inhibited the experssion of IL-6, ABP, vimention and pan-cadherin on Sertoli Cells of male SD rats in sexual maturation period.
