外置复合磁场导引下铁碳复合磁性药物载体在兔肝脏的靶向分布

摘  要:目的应用复合磁场导引和肝动脉介入插管方法,实现铁碳复合磁性载体在兔肝靶区的靶向定位。方法制备出永磁场与脉冲磁场相结合的复合磁场并测定复合后的磁感应曲线。12只新西兰白兔随机分成3组,每组4只:①靶向组:将吸附Na99mTc O4的铁碳复合载体注入兔左肝动脉,左肝相应体表置复合磁场。②非靶向组:将吸附Na99mTc O4的铁碳复合载体注入兔左肝动脉,无外置磁场。③对照组:兔左肝动脉注入Na99mTc O4水溶液。3组分别于注药后15min、30min、1h、2h、3h、4h进行SPECT显像。兔左肝作组织切片观察。结果复合磁场具有更强的磁感应强度;靶向组在注药后各时段的肝感兴趣区每像素放射计数均显著高于非靶向组及对照组(P<0.05);铁碳复合磁性载体分布于靶向组的肝细胞浆及组织间质内,非靶向组仅分布于肝血窦内。结论在外置复合磁场引导下铁碳复合磁性药物载体通过介入途径能有效实现在兔肝脏的靶向分布。Purpose To study the targeted distribution of magnetic drug carrier of iron-carbon composite particles in rabbit liver by application of external complex magnetic field and artery cannulation. Methods A new complex magnetic field combined permanent magnet with pulse magnetic emitter was developed and its magnetic induction curve was measured. Twelve NZW rabbits were randomly assigned to 3 groups, each group had 4 rabbits. ①targeting group: Na^99mTcO4 iron-carbon composite particles were injected into the left hepatic artery of rabbit and the external complex magnetic field was placed over the surface of the left liver. ②Non-targeting group: Na^99mTcO4 iron-carbon composite particles were injected into the left hepatic artery of rabbit without external magnetic field. ③control group: Na^99mTcO4 saline was directly injected into the left hepatic artery of rabbit. Each rabbit received SPECT scintigraphy at 15 min,30 min,1 h,2 h,3 h,4 h after injection. Each rabbit was sacrificed and its left liver was histologically studied after SPECT imaging. Results Complex magnetic field showed significantly improved property of magnetic induction. Targeting group had higher counts/ pix of liver ROI than the other two groups in each time span, the difference was statistically significant (P〈0.05). Magnetic iron-carbon composite particles were distributed in rabbitrs liver cell plasma and matrix in targeting group, while they were only existed in liver sinusoid in non-targeting group. Condusions Under the direction of external complex magnetic field, magnetic iron-carbon drug carrier can be effectively targeted at rabbitls liver by interventional approach.
