摘 要:目的观察阿司匹林对高脂性动脉粥样硬化兔血脂和血小板聚集的影响以及两者之间的相互关系。方法对雄性新西兰兔给予高脂饮食加腹主动脉内膜剥脱术制成高脂性动脉粥样硬化模型,然后采用阿司匹林5~20 mg/kg治疗4周,用比色法测定各组兔的血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白-胆固醇(HDL-C);用比浊法测定ADP和花生四烯酸诱导的血小板聚集性。结果阿司匹林治疗4周后,血清TC降低25.7%~48.0%,低密度脂蛋白(LDL-C)降低38.3%~66.2%,HDL-C升高33.3%~70.8%,LDL-C与HDL-C比值降低48.3%~81.9%。阿司匹林(5~10mg/kg^(- 1))对ADP或花生四烯酸诱导的血小板聚集均有较好的抑制作用,当剂量增加至20mg/kg^(-1)时,对血小板的聚集抑制作用反而减弱。结论阿司匹林对高脂性动脉粥样硬化兔具有明显的调脂和抑制血小板聚集的作用,但两者发挥最佳作用的剂量不同。Objective To observe the effect of aspirin on blood lipid and platelet aggregation in atherosclerotic rabbits with hyperlipidemia, and the relationship between them. Methods The hyperlipidemic atherosclerotic model was developed with male New Zealand rabbits that were given high fat diet and abraded of the abdominal aorta endothelial cells. These rabbits were then administered with 5 to 20 mg of aspirin per kg of body weight for 4 weeks, serum total cholesterol(TC), triglyceride(TG), high density lipoprotein-cholesterol(HDL-C) were measured with colorimetric method; platelet aggregation induced by ADP and arachidonic acid was determined with turbidimetric method. Results After the rabbits were treated with 5-20mg/kg aspirin for 4 weeks, serum TC was decreased by 25.7%-48.0%, low density lipoprotein-cholesterol (LDL-C) 38.3%-66.2%, HDL-C was increased by 33.3%-70.8%, and the LDL-C/HDL-C ratio was decreased by 48.3%- 81.9 %. The treatment of 5 to 10 mg of aspirin per kg of body weight could inhibit platelet aggregation induced by ADP or arachidonic acid, but the inhibition at a dose of 20 mg of aspirin per kg of body weight against platelet aggregation was weakened in- versely. Conclusion Aspirin not only obviously regulated blood lipid but also evidently inhibites platelet aggregation in atherosclerotic rabbits with hyperlipidemia, however, the dosage at which aspirin exerted the best effect on these two factors was different.
