摘  要:目的探讨电学环境对培养在聚吡咯/聚乳酸(PPy/PLA)骨组织工程材料上成骨细胞的影响。方法化学氧化法制备含有10%PPy的PPy/PLA复合膜,将3~6代原代培养的成骨细胞种植其上,使用0、12.5、25、50、75、100、125、150、175和200μA/cm2的电流密度刺激,分别使用MTT法、碱性磷酸酶(ALP)活性和胞外钙基质的含量测定成骨细胞在复合膜上的增殖、分化和矿化情况。结果50和75μA/cm2是促进细胞增殖的最适合电流值,50μA/cm2能显著促进细胞的分化和矿化。结论电刺激可以促进种植在电活性材料上成骨细胞的增殖,提高细胞的活性,为骨组织的重建、加速骨界面的愈合开辟一条新的道路。Objective To discuss the effect of electro-circumstance on osteoblasts which were cultured on the PPy/PLA bone tissue engineering materials. Methods 3 -6 generation osteoblasts were implanted on PPy/PLA electroactive composite membranes containing 10% PPy which was prepared by chemical oxidation. Then ten levels electrical currents ,0,12.5,25,75,100,125,150,175 and 200 μA/cm^2 , were used to stimulate the osteoblasts . Proliferation, differentiation and mineralization were measured by MTT method,ALP activity and extracellular calcium matrix of the cells respectively. Results The eletrieal currents of 50 and 75μ A/cm^2 were more suitable electrical currents than others for the cell proliferation. 50μA/cm^2 eletrieal current can obviously accelerate differentiation and mineralization. Conclusions Electrical stimulation promotes the proliferation of osteoblasts implanted on the electro-active materials proliferation and their activities. This will be a new road for bone tissue rebuilding and accelerate bone interfaeial healing up.
