摘 要:为探讨鸡胚心脏发育过程中细胞增殖的变化情况,采用免疫组织化学SP法和AgNOR银染法分别检测了第7~18天鸡胚心脏PCNA和AgNOR的分布。结果显示:1)PCNA免疫反应阳性心肌细胞主要分布于心包侧,随胚龄增加阳性率下降,至第13天后呈小幅回升,第7~18天平均阳性率为29.20%;PCNA免疫反应阳性内皮细胞在胚胎早期分布于心内膜内皮,胚胎中后期主要为心肌血管内皮呈阳性,第7~13天,内皮处于低增殖阶段,第14天后逐渐增加,第7~18天平均阳性率为15.33%。2)AgNOR阳性心肌细胞第7天为聚集型,第8~14天为混合型,第15~18天为单一型,随胚龄增加阳性率下降,至第15天后呈小幅回升,第7~18天平均阳性率为58.84%;AgNOR阳性内皮细胞类型不易区分。第7~9天平均阳性率为9.25%,此后阳性率逐渐上升,至第18天时达到最高为27.07%,第7~18天平均阳性率为13.95%。结果表明,第7~18天鸡胚心脏发育过程中以心肌细胞增殖为主,内皮细胞增殖较慢且较心肌细胞滞后。第15天后心肌细胞和内皮细胞增值率均升高,可能与心肌-内皮通过某些信号途径相互作用有关。This study examined the distribution of PCNA and AgNOR in the heart of chick embryo aged 7 to 18 days,using SP Immunohistochemical and AgNOR silver staining methods to determine changes in cell proliferation during development. The results were as follows: 1) PCNA immunoreactive positive cardiac muscle cells were mainly distributed in the pericardia. The percentage of PCNA positive cells decreased with development, but increased slightly after E13. The average positive percentage was 29.20% from E7 to E18. The PCNA immunoreactive positive endothelial cells were distributed in the endocardium at the middle and late stages of the embryo and in blood vessels of the cardiac muscle at the laters stage of the embryo. The percentage of cell proliferation from E7 to E13 was low but increased thereafter. The average positive percentage was 15.33% from E7 to E18. 2) The AgNOR positive cardiac muscle cells were in an aggregated Form at E7, Mixed Form at E8 to E14 and Single Form at E15 to E18. The percentage of AgNOR positive cells decreased with development, but increased slightly after E15. The average positive percentage was 58.84% from E7 to E18. Types of AgNOR positive endothelial cells were not distinguishable. The average positive percentage was 9.25% from E7 to E9. Thereafter, the positive percentage increased gradually up to 27.07% at E18. The average positive percentage was 13.95% from E7 to E18. In conclusion, the study indicated that the main proliferation cells were cardiac muscle cells and the proliferation of endothelial cells was slower and later than cardiac muscle cells during the development of chick embryo heart from E7 to E18. The increase in proliferation percentage of both cardiac muscle cells and endothelial cells after E15 may be related to the signal pathway between the cardiac muscle and endothelium.
