单色光对肉鸡甲状腺形态结构及PCNA表达的影响

摘  要:本试验以LED灯(发光二级管)为光源,比较单色光对AA(爱拔益加)肉鸡甲状腺组织形态结构以及细胞增殖核抗原(PCNA)表达的影响。将60羽刚出壳的雄性AA肉鸡分别饲养在红光(660nm)、绿光(560nm)、蓝光(480nm)和白光(400~760nm,对照组)下49d(n=15),光照度15lx,光照制度(L/D)23h/1h。取甲状腺,做石蜡切片,HE染色测量甲状腺滤泡面积和滤泡上皮细胞高度及PCNA免疫组化染色阳性细胞表达率。结果显示:红光组肉鸡甲状腺绝对质量、甲状腺滤泡面积比其他光组分别低4.67%~41.47%和22.16%~50.32%(P〈0.05),而甲状腺相对质量、滤泡上皮细胞高度和PCNA阳性细胞表达率比其他光组分别高7.98%~22.97%、11.92%~26.72%和65.25%~89.09%,且组间差异显著(P〈0.05)。结果表明:红光(660nm)能促进甲状腺滤泡上皮细胞的生长和PCNA阳性表达率。The effect of monochromatic light on morphology of thyroid and expression of proliferating cell nuclear antigen (PCNA) were studied in AA (Arbor Acres) broilers exposed to red (660 nm), green (560 nm), blue (480 nm) and white light (400-760 nm, control) using LED (light-emitting diodes) as light sources for 49 d,respectivdy. Light intensity was 15 lx and schedule was 23L : 1D during the entire experiment. The thyroids were weighed, sectioned and stained with HE and PCNA immunohistochemical. The follicle area and height of epithelial cells well as the percentage of PCNA positive cells were measured in the thyroid, The results showed that absolute weight and follicle area of thyroid were lower in the red light group than in other light groups (4.67% -41.47% and 22.16% -50.32%, P〈0.05). However, the thyroid relative weight, height of thyroid epithelial cells and percentage of PCNA positive cells were higher in the red light group than in other light groups (7.98% -22.97%, 11.92% -26. 72% and 65.25% - 89.09%, P〈0.05). This study indicates that the red light (660 nm) stimulated the growth of epithelial cells and expression of PCNA in the thyroid of broilers.
