摘 要:目的制备一种新型骨髓干细胞富集材料,并观察其表征。方法用预先制备的人脱钙骨基质(DBM)与多聚左旋赖氨酸(PLL)复合制备富集材料。参照Zhang氏液体置换法测定其密度与孔隙率;三维视频显微镜、扫描电镜观察其孔径、表面及横断面超微结构;拉曼光谱分析鉴定材料成分,并进行组织学观察。结果经PLL修饰的DBM富集材料(PLL-DBM)密度为(0.27±0.02)g/ml,孔隙率为(73±11)%,孔径为(412.73±160.29)μm。孔隙内部有大量孔隙相互连通,PLL在材料内外表面形成均匀的乳白色涂层,并在天然孔隙内形成更小、孔径较均匀的网孔结构。结论制备的PLL-DBM具有自体骨三维空间结构和促进细胞粘附的PLL,是一种较理想的骨髓干细胞富集材料。Objective To prepare and measure a new kind of scaffold for enriching bone marrow stem cells by selective cell retention. Methods Decalcified bone matrix (DBM) was decorated with poly-L-Lysine (PLL) as the scaffold material (PLL-DBM). Porosity and density were measured with Zhang's process, Pore size, surface and cross section were observed under three-dimension video microscope and scanning electron microscope. Material component were analysized by Raman spectrum and histological observation. Results The density, porosity and pore size of PLL-DBM were (0. 27± 0. 02)g/ml, (73 ± 11)% and (412.73 ± 160.29)μm respectively. DBM were covered with uniformly-distributed and ivory white PLL, The PLL-DBM had high porosity with much more interior cross-connection pore. Conclusion The new constructed PLL-DBM has as good three-dimension space as autogenous bone, and well decorated with PLL for facilitating cells adhesion. It is an effective material for enriching bone marrow stem cells.
