摘 要:目的:探讨胃肠舒和血流干预药物对实验性糖尿病和糖尿病胃轻瘫大鼠的影响。方法:131只SPF级SD雄性大鼠,随机分为正常组(10)、模型组(22)、止血敏组(33)、蚓激酶组(33)和胃肠舒组(33),除正常组外大鼠一次性腹腔注射链脲佐菌素(60mg/kg)造模,7天后以血糖≥16.7mmol/L为实验纳入标准。造模10天后,胃肠舒组予胃肠舒灌胃,止血敏组予止血敏灌胃,蚓激酶组予蚓激酶腹腔注射,模型组和正常组予等量蒸馏水灌胃,每日1次。30周后检测血糖、FINS、C-P、TC、TG、LDL、HDL、胃液体排空率等指标;并取胰腺组织,光镜下观察胰岛和胰岛β细胞变化。取胃窦组织,光镜下检测胃窦肌间神经丛中nNOS免疫反应性神经元含量。结果:30周后造模组大鼠血糖、FINS、C-P、及血脂的变化与正常组相比均有显著性差异(P<0.01),胰腺观察到胰岛β细胞减少,有的甚至消失;而胃肠舒组各项生化指标较模型组有改善,胰腺见血管充血现象较模型组减轻,胰岛β细胞明显增多。胃液体排空率,胃肠舒组明显高于模型组、蚓激酶组及止血敏组(P<0.01);胃窦肌间神经丛中nNOS染色阳性神经含量,胃肠舒组也显著多于模型组、蚓激酶组及止血敏组(P<0.01)。结论:胃肠舒具有降血糖和促进胃排空双重功效。血流干预药物对大鼠胃液体排空有一定影响。Objective : To investigate the effect of medicine interfering in blood flow and "Weichangshu" ( WCS ) to rat' s diabetic model and rat' s diabetic gastroparesis model. Methods : One hundred thirty-one male SD rats who were SPF class were randomly divided into the control group ( 10 rats ), model group ( 22 rats ), ethamsylate group ( 33 rats ), lumbrukinase group ( 33 rats ) and WCS group ( 33 rats ) .Except for rats in the control group, surplus rats were injected streptozotocin ( STZ ) solution ( 60mg/kg ) into their abdominal cavities. We measured blood sugar after seven days making diabetic mould and looked blood sugar more than or equal to 16.7mmol/L as experimental internalized standard. After 10 days, rats in the WCS group were treated by traditional Chinese medicine "WCS" apozem which were given by intragastric adminisreation one time per day. Rats in the ethamsylate group were treated by ethamsylate injection which were given by intragastric adminisreation one time per day. Rats in the lumbrukinase group were treated by lumbrukinase enteric-coated capsules which were injected into their abdominal cavities one time per day. Rats in the control group and rats in the model group were treated by tales doses distilled water which were given by intragastric adminisreation one time per day. After having been observed for 30 weeks, all rats were measured contents of blood sugar, FINS, C-P, TC, TG, HDL, LDL, rats' gastric fluid emptying rate. Subsequently, we got out the stomach and pancreas, and knew how pancreatic tissue and pancreatic islet were destructed, and observed pathological lesion of gastric neurotransmitter such as nNOS through light microscope. Results : after 30 weeks, blood sugar, FINS, C-P, TC, TG, HDL and LDL in rats of all made diabetic model group were significantly difference compared with ones in rats of the control group (P〈 0.01 ), and there were no or few beta cells in rats' pancreatic islets.
