摘  要:目的:通过在培养基中添加葡萄糖的方法,提高转基因鱼腥藻的产量和人肿瘤坏死因子(hTNF)-α的表达率。方法:在葡萄糖浓度为0~300mmol/L的范围内,进行了转hTNF-α鱼腥藻IB02的摇瓶混合营养培养,用比浊法和酶联免疫法测定转基因鱼腥藻的生长和hTNF-α的表达。结果:添加葡萄糖的藻液最高生长密度是未添加葡萄糖的3.5倍,且hTNF-α的表达率也提高至4倍。结论:在各种葡萄糖浓度下,葡萄糖的利用都不明显。Objective: To improve the biomass of transgenic Anabaena sp. PCC IB02 harboring human tumor necrosis factor-a(hTNF-a) gene and the expression efficience of hTNF-a by the method of adding the glucose to the BG-11(-N) medium. Methods: Mixotrophic cultivation was carried out in flasks with glucose concentrations in the range from 0 to 300 mmol/L in BG-11(-N) medium. The turbidimetric method and enzyme-linked immunosorbent assay(ELISA) method were used to measure the biomass and the expression efficience of hTNF-a. Results: The data showed that the cell density increased to 3.5 times of that in control sample, and the expression rate of hTNF-a increased to 2%, from 0.5% in no glucose. Conclusion: At different glucose concentrations, the glucose consumption was not obviously.
