摘 要:目的研究红景天甙对帕金森病(PD)小鼠脑组织中PI3K/蛋白激酶B(PKB)信号转导途径的影响及其神经保护作用的机制。方法30只C57BL雄性小鼠随机分为PD组、红景天甙组和正常对照组,每组10只。用蛋白印迹法检测小鼠纹状体中Akt/pAkt(ser473)及其下游糖原合成酶激酶-3β(GSK-3β)/磷酸化糖原合成酶激酶-3β[pGSK-3β(ser9)]、天冬氨酸特异性半胱氨酸蛋白酶3(Caspase-3)、BAD和Bcl-2的表达。结果红景天甙组及正常对照组纹状体pAkt(ser473)表达(0.5487±0.01376,0.5055±0.0524)比PD组(0.3069±0.08734)显著增高(均P<0.05);pGSK-3β(ser9)蛋白表达(0.6959±0.3327)较PD组(0.4884±0.02257)显著增高(P<0.05)。红景天甙组的Bcl-2/BAD比值(1.2669±0.231)明显高于PD组(0.4583±0.088)(P<0.05)。红景天甙组小鼠纹状体中活化的Caspase-3蛋白(0.0874±0.0109)比PD组(0.1409±0.089)明显下降(P<0.05)。结论红景天甙能使PI3K/PKB信号转导途径中Akt在ser473位点以及GSK-3β在ser9位点的磷酸化程度增高、上调Bcl-2/BAD比值、抑制Caspase-3蛋白的活化,发挥神经保护作用。Objective To investigate the effect of Salidroside on PBK/protein kinase B (PKB) signaling in Parkinson's disease(PD) model mice and its mechanism of neuroprotection. Methods The 30 C57BL mice were divided into 3 groups: PD group(10), Salidroside group(10) and normal control(NC) group(10). The protein expressions of Akt, pAkt( ser 473), GSK-3β, pGSK-3β (ser 9), Bcl-2, BAD and Caspase-3 were detected by Western blot. Results The protein levels of pAkt ( ser 473) in striatum were significantly increased in Salidroside group (0. 5487 ±0. 01376) and NC group ( 0. 5055 ± 0. 0524 ) than that in PD group ( 0. 3069 ± 0. 08734 ) ( all P 〈 0. 05 ). The expression of pGSK-3β ( ser 9 ) protein was obviously increased in Salidroside group (0. 6959 ± 0. 0332 ) than that in PD group ( 0. 4884 ±0. 0225 ) ( P 〈 0. 05 ). The BcI-2/BAD ratio of Salidroside group ( 1. 2669 ±0.231) was much more increased than that of PD group (0. 4583 s0. 088) (P〈0. 05). Expression of active Caspase-3 in Salidroside group(0. 0874 ± 0. 0109 ) was much lower than that in PD group (0. 1409 ± 0. 089 ) (P 〈 0. 05 ). Conclusions The Salidroside can increase phospholateion activation of Akt at ser 473 site and GSK-3β at ser 9 site in PI3K/PKB, can partially increase the Bcl-2/BAD ratio and inhibit the activation of Caspase-3 protein, therefore it can play neuroprotective effect.
