摘 要:目的研究镍冶炼烟尘对NIH/3T3细胞的毒性。方法以我国某镍冶炼厂的镍冶炼炉前沉降尘和镍精矿车间沉降尘为受试物,观察NIH/3T3细胞对其的吞噬活性,并用噻唑蓝(MTT)比色的方法检测细胞活性。结果两受试物均可被NIH/3T3细胞吞噬,在200μg/ml时的吞噬率分别为100.0%、83.0%;100μg/ml时的吞噬率为77.0%、58.8%;50μg/ml时的吞噬率为59.8%、30.5%,差异均有统计学意义(P<0.05);且随作用时间增长,细胞生长受到抑制程度也随之增加。接触24h,两受试物细胞半数抑制浓度分别为19.86、30.15μg/ml。结论两受试物均可诱导NIH/3T3细胞吞噬活性,可以降低细胞的成活率,为镍冶炼烟尘的致癌危险性以及镍冶炼工肺癌病因学研究提供新的实验依据。Objective To study the toxicity of nickel-smelting fume on NIH/3T3 cells. Methods To observe the effect of two kinds of nickel fumes collected from smelting furnace and refining workshop of a nickel smeltery in China on the phagocytizing activity of NIH/3T3 cells; meanwhile, the cytoactivity was also detected with MTT colorimetric assay. Results It was shown that two kinds of nickel fumes all could be phagocytizied by NIH/3T3 cells. The phagocytizing rate was dose-dependent : at the exposed level of 200μg/ml, the rates were 100. 0% and 83.0% ; and 77.0%, 58. 8% at 100μg/ml; and 59.8%, 30. 5% at 50μg/ml, respectively, there were significant differences. Additionally, with the extension of exposure time, the growth of NIH/ 3T3 cells was also inhibited. After 24 hours exposure, the 50% inhibiting concentrations of cell growth were 19. 86 and 30, 15μg/ml respectively for these two kinds of fumes. Conclusions Two kinds of the fumes all might induce the phagocytizing activity and depress the survival rate of NIH/3T3 ceils. These findings provide a new experimental evidence for the carcinogenic risk of nickel-smelting fume and the etiological agent of lung cancer in nickel-smeltlng workers.
