摘  要:目的研究Alzheimer相关的神经丝蛋白(Alzheimer-associated neuronal thread protein,AD7C-NTP)在老年痴呆(Alzheimer’s disease,AD)患者脑内、猪脑和小鼠脑内的表达,初步确定一种AD7C-NTP蛋白含量丰富的组织来源。方法采用免疫组织化学染色方法观察猪脑、小鼠脑内AD7C-NTP的分布,采用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)和蛋白印迹(Western blot)的方法检测AD患者脑内、猪脑、小鼠脑内AD7C-NTP的表达水平。结果免疫组化结果显示,猪大脑皮质、小鼠海马内有AD7C-NTP免疫反应阳性神经元分布;Western blot的结果显示,以AD7C-NTP C端抗体作为检测抗体时,AD患者脑和猪脑内可见相对分子质量为41000bp的免疫反应阳性条带,小鼠脑内未见免疫反应阳性条带。结论AD患者脑内、猪脑内存在AD7C-NTP,可作为提取AD7C-NTP蛋白纯品的来源之一,为研制AD7C-NTP诊断试剂盒提供实验材料。Objective To investigate the expression of Alzheimer-associated neuronal thread protein(AD7C-NTP) in the brain of patients with Alzheimer's disease(AD), as well as in swine and mice, so as to determine the source of tissues rich in AD7C-NTP. Methods The distribution of AD7C-NTP in the brain of pig and mouse was investigated with immunohistochemical methods. SDS-PAGE and Western blot were applied to detect the expression of AD7C-NTP in the brain of patients with AD as well as in pigs and mice. Results The results of immunohistochemistry indicated that AD7C-NTP immunoreactive positive neurons were observed in the cerebral cortices of pigs and hippocampi of mice. The results of Western blot indicated that a band of about 41 000 bp was detected with the antibody of AD7C-NTP against C-terminal in the brain of patients with AD and in pigs. Conclusion The existence of AD7C-NTP in the brain of patients with AD is confirmed; AD7C-NTP is also found in the brain of swine, so that it can be considered as a source for extraction of purified AD7C-NTP protein, which can be used as the experimental material for AD7C-NTP diagnostic kit.
