摘 要:目的对掌叶大黄乙醇提取物中天然成份-蒽醌类化合物的抑制乙肝病毒(HBV)的活性进行检测。方法制备掌叶大黄乙醇提取物(RPE)并从中分离提取有效成份,荧光定量PCR法和ELISA法检测PRE及其各成份对Hep-AD38细胞中HBV表达及复制的作用;同时用MTT法检测细胞毒性。结果从掌叶大黄乙醇提取物中分离得到的大黄酚-8-O-β-D-葡萄糖甙对HBV DNA复制以及抗原表达均有明显的抑制活性。同时MTT法检测显示大黄酚-8-O-β-D-葡萄糖甙没有明显毒副作用。结论大黄酚-8-O-β-D-葡萄糖甙可以有效抑制HBV DNA复制及HBsAg表达,对其进一步的研究将有助于在此化合物基础上开发出新的抗HBV药物。Objective To analyze the inhibitory effect of anthraquinones isolated from traditional Chinese medicine Rheum palmatum L. on HBV-DNA replication and HBV antigen expression in a stable HBV-produeing cell line HepAD38. Methods The Rheum palmatum L. ethanol extract (RPE) was prepared and the active compounds from RPE were isolated by RP-HPLC. Quantitative PCR analysis and ELISA were employed to analyze the inhibitory effect of RPE and the compounds. The cell growth inhibiting effects of RPE and the compounds were determined with MTT assay. Resuhs From six anthraquinones isolated from RPE, the only combined anthraquinone ehrysophanol 8-O-β-D-glueoside exhibited significant effects against HBV DNA production and antigen expression. Moreover, the ehrysophanol 8-O-13-D-glueoside did not show significant eytotoxieity. Conclusion The ehrysophanol 8-O-β-D-glueoside is the active compound from Rheum palmatum L which can effectively in hibite HBV DNA replication and HBsAg expression.
