摘  要:目的:探讨丙酮酸氧化酶法在测定血清丙氨酸氨基转移酶中的作用.方法:利用丙酮酸氧化酶氧化丙酮酸生成过氧化氢,过氧化氢与2,4,6三溴-3-羟基苯甲酸和4-氨基比林在过氧化物酶作用下生成红色醌类化合物的原理.结果:该方法线性范围达200 U/L,与紫外酶偶联速率法比较,相关系数r=0.989,y=1.075x+1.031,n=80.结论:该方法具有线性范围大、灵敏、操作简便、快速、结果准确、重复性好、抗干扰能力强等优点,适合血站大批量标本的检测.Objective: To investigate the application of pyruvate oxidase method to detect serum ALT with enzyme labeling meter. Methods: On the basis of the principle of pyruvate oxidase oxygenizing pyruvate to form hydrogen dioxide, and hydrogen dioxide combined with TBHBA and 4 - AAP to form red quinone compound catalyzed by peroxydase. Results: The linear range of this method amounted to 200U/L, and coefficient correlation of this method was r = 0. 989, y = 1. 075x + 1. 031 (n = 80)compared with UV method of coupling method. Conclusion: The method possesses the merits of wide linear range, sensitiveness, simple manipulation, high - speed, accuracy, good reproducibility and great ability of antiturbulence. It is suitable to detect the enormous blood samples in blood bank.
