摘  要:目的:观察大黄虫丸对大鼠脑出血模型脑组织IL-1βmRNA、iNOSmRNA表达的影响。方法:以雄性Sprauge-Dawley大鼠为实验动物,共分4组,正常组、假手术组、模型组、中药组。模型组和中药组选择胶原酶加肝素联合注射法诱导脑出血大鼠模型,中药组予大鼠大黄虫丸的混悬液2.5ml灌胃,每天1次,正常组、假手术组、模型组予等量的生理盐水灌胃,每天1次,3天后将大鼠断头处死,快速取出血肿周围组织约100mg,以RT-PCR法测定脑组织中IL-1βmRNA、iNOSmRNA表达。结果:正常组、假手术组脑组织中IL-1βmRNA、iNOSmRNA仅有少量表达,模型组、中药组组织的IL-1βmRNA、iNOSmRNA表达明显上调,和正常组、假手术组有明显的差异(P<0.05)。但中药大黄虫丸能明显的下调脑出血大鼠脑组织IL-1βmRNA、iNOSmRNA的表达,和模型组相比(P<0.05)。结论:胶原酶加肝素联合注射法诱导脑出血大鼠模型血肿周围脑组织IL-1βmRNA、iNOSmRNA表达增高,大黄虫丸对脑出血大鼠脑组织IL-1βmRNA、iNOSmRNA的表达增加有下调作用。objective: To study the effect of Dahuangzhechong pill on expression of IL-1βmRNA and iNOS mRNA in brain tissue of cerebral hemorrhage rat model. Methods: Male Sprauge-Dawley rats were divided into normal group, model group, sham operation group and therapy group. The model was established by injecting eollagenase and heparin. 2.5ml Dahuangzheehong mixture was given to each rat in therapy group once daily, while N. S. of the same volume was given to other groups. After 3 days, all rats were saeri- riced hy deeollation and 100mg peri-hematoma tissue was taken immediately. The expression of IL-1βmRNA and iNOS mRNA was examined through RT-PCR. Results: The expression of IL-1βmRNA and iNOS mRNA in brain tissue in control group and sham group was higher than that in model group and therapy group ( P 〈 0.05 ), and in therapy group it was lower than that in model group ( P 〈 0.05). Conclusion: The high expression of IL-1βmRNA and iNOS mRNA in brain tissue could be caused by cerebral hemorrhage, and Dahuangzheehong pill could decrease it.
