摘  要:研究曲尼司特对环孢素A(CsA)慢性肾毒性大鼠的肾脏保护作用,发现曲尼司特能减低环孢素A慢性肾毒性大鼠血浆和肾组织中AngⅡ的含量,并下调肾组织中ET-1的表达.结果表明曲尼司特可能通过下调环孢素A慢性肾毒性大鼠血浆和肾组织中AngⅡ的的水平以及肾组织中ET-1的表达,减轻肾脏病理损害,发挥其肾脏保护作用.To investigate the effects of tranilast, the anti-allergic agent N-(3,4-dimethoxyciannamoyl)-anthranilic acid, on angiotensin Ⅱ and endothelin-1 in rats with chronic cyclosporin A nephrotoxicity. It was showed: The expression of Ang Ⅱ in the plasma and cortex of kidneys in group M showed significantly higher than it in group N (489.22± 9.79 versus 420.21 ± 9.36× 10^- 12 g/mL; 82.72 ± 8.69 versus 40.74 ± 5.32 × 10^-12 g/mg tissue). Like irbesartan(457.69 ± 5. 13 × 10^-12 g/mL; 53.93 ± 5.44 × 10^-12 g/mg tissue), Tranilast significantly decreased the level of Ang Ⅱ in plasma (466.25 ±16.87 × 10^-12 g/mL) and kidney tissues(62.25 ± 14.31 × 10^-12 g/mg tissue). Morphologically, kidneys in group M exhibited significantly more fibrosis, which was blunted by tranilast. In addition, tranilast decreased the expression of ET-1 in kidneys of rats with chronic cyclosporin A nephrotoxicity. Tranilast showed a renal protective effect on chronic cyclosporine A nephrotoxicity of rat model by decreasing the level of endothelin-1 and angiotensin Ⅱ.
