菊藤胶囊对应激大鼠血浆及肾上腺皮质血管紧张素Ⅱ水平的影响

摘　　要：目的探讨中药复方菊藤胶囊对慢性应激性高血压大鼠血压、血浆及肾上腺组织血管紧张素Ⅱ(AngⅡ)的影响。方法大鼠随机分为模型组、菊藤胶囊高剂量组、菊藤胶囊中剂量组、菊藤胶囊低剂量组、卡托普利组、正常对照组;除正常对照组外,其余五组均采用低频低压交流电间断电击法。电击大鼠足底28d,制作应激性高血压模型,同时各组均加入不同的干预。应用经尾动脉测量血压和心率,酶联免疫吸附法检测大鼠血浆AngⅡ,免疫组织化学法检测肾上腺AngⅡ。观察菊藤胶囊不同剂量组、卡托普利组对应激致大鼠血压及相关指标的影响。结果与正常对照组相比,模型组大鼠血压和血浆AngⅡ含量明显升高(P<0.01),AngⅡ在肾上腺皮质表达增强(P<0.01);与模型组相比,菊藤胶囊高、中剂量组均能抑制应激大鼠的血压及血浆AngⅡ含量的升高(P<0.01),同时降低AngⅡ在肾上腺皮质的表达(P<0.01)。结论菊藤胶囊能够降低应激性高血压大鼠的血压,其机理可能是通过抑制血浆及肾上腺组织中AngⅡ的含量的升高实现的。Objective： To study the effect of Juteng capsule on blood pressure, plasma and adrenal Ang Ⅱ of stress - induced hypertension in rats. Methods： 42 healthy male SD rats were randomly divided into 6 groups. They were model group, JT - H group, JT - M group, JT - L group, captopril group and control group. Electric foot shock was used to build stress - induced hypertension rats. Observed the blood pressure, level of plasma and adrenal cortex Ang Ⅱ in rats. The level of plasma Ang Ⅱ were measured by ELISA. The level of adrenal cortex Ang Ⅱ were measured by SP. Results： Compared with control group, the blood pressure, plasma Ang Ⅱ content and Ang Ⅱ expression in adrenal cortex of model group were significantly higher. Compared with model group, they were significantly lower than JT - H, JT - M and captopril. Conclusion： Juteng capsule can significantly degrade the blood pressure, perhaps by degrading plasma and adrenal cortex Ang Ⅱ.
